平贝母多糖的提取工艺及其体外抗氧化活性研究 被引量:2

Study on Extraction Technology of Polysaccharides from Fritillaria ussuriensis and Its Antioxidant Activity

摘要 优化平贝母多糖的提取工艺并对其体外抗氧化活性进行研究,为进一步开发利用平贝母资源奠定基础。利用超声提取方法,采用响应面法优化平贝母多糖的提取工艺,通过测定平贝母多糖对DPPH自由基的清除率,评价其体外抗氧化活性。结果表明,平贝母多糖最佳提取工艺是料液比1︰29 g/mL,超声功率50 W,超声时间14 min,提取温度34℃。平贝母多糖的提取率为9.70%±0.10%。平贝母多糖主要由Glc组成,含有少量Ara和Gal,摩尔比99.3︰0.3︰0.4。平贝母多糖对DPPH自由基具有较好的清除能力,平贝母多糖质量浓度4 mg/mL时,其对DPPH自由基的清除率为60.40%。响应面法优化超声提取平贝母多糖工艺是可行的,平贝母多糖具有较好的体外抗氧化能力,为平贝母多糖产品开发及利用提供试验依据。 To optimize the extraction process of polysaccharides from Fritillaria ussuriensis,its antioxidant activity was evaluated,which could provide a theoretical support for its further development and utilization.The polysaccharide of Fritillaria ussuriensis was extracted by ultrasonic extraction method.The extraction process of polysaccharides from Fritillaria ussuriensis was optimized by response surface analysis,at the same time,the scavenging capacity of Fritillaria ussuriensis polysaccharides to DPPH free radical was determined.The optimum condition for the extraction was as follows:liquid to material ratio 1︰29 g/mL,ultrasonic power 50 W,ultrasonic time 14 min,and extraction temperature of 34℃.Under this condition,the extraction rate of polysaccharides from Fritillaria ussuriensis was 9.70%±0.10%.The results showed that the polysaccharide of Fritillaria ussuriensis was a pyran ring structure,consisting of glucose and a small amount of arabinose and galactose in the molar ratio of 99.3︰0.3︰0.4.Fritillaria ussuriensis polysaccharides had a certain scavenging effect on DPPH free radical.When the concentration was 4 mg/mL,the DPPH clearance rate was 60.40%.The parameters of polysaccharides extraction process optimized by response surface method were feasible.Fritillaria ussuriensis polysaccharides had good antioxidant activity in vitro.It provided an experimental basis for the further development and utilization of the polysaccharides from Fritillaria ussuriensis.
