HPLC波长切换法同时测定栀子柏皮汤中8种成分的含量 被引量:1

摘要 目的建立高效液相色谱(HPLC)波长切换法同时测定栀子柏皮汤中8种成分京尼平龙胆双糖苷、栀子苷、盐酸黄柏碱、4-O-阿魏酰奎尼酸、5-O-阿魏酰奎尼酸、甘草苷、盐酸小檗碱和甘草酸的含量。方法色谱柱采用Phenomenex Kinetex C18柱(150 mm×4.6 mm,2.6μm);流动相为乙腈-0.05 mol·L^(-1)磷酸二氢钾溶液(磷酸调pH至3.0)进行梯度洗脱;流速为1.0 mL·min^(-1);柱温40℃;检测波长为240 nm(检测京尼平龙胆双糖苷、栀子苷、盐酸黄柏碱)、280 nm(检测4-O-阿魏酰奎尼酸、5-O-阿魏酰奎尼酸、甘草苷、盐酸小檗碱)和250 nm(检测甘草酸)。结果京尼平龙胆双糖苷、栀子苷、盐酸黄柏碱、4-O-阿魏酰奎尼酸、5-O-阿魏酰奎尼酸、甘草苷、盐酸小檗碱和甘草酸分别在质量浓度15.57~185.9、59.18~706.6、5.316~63.48、14.10~168.3、11.92~142.4、3.078~36.76、20.57~245.6、6.150~73.43 mg·L^(-1)内与峰面积呈良好的线性关系(r>0.999),平均加样回收率分别为102.1%、99.7%、98.3%、94.8%、99.5%、99.6%、98.2%和103.5%,RSD均小于2%(n=6)。结论本研究建立的HPLC波长切换法简便可行,可用于栀子柏皮汤中上述8种成分含量的同时测定。 Objective To establish a high performance liquid chromatography(HPLC)wavelength switching method for simultaneous determination of 8 components including genipin gentiobioside,geniposide,phellodendrine chloride,4-O-feruloylquinic acid,5-O-feruloylquinic acid,liquiritin,berberine hydrochloride and glycyrrhizic acid in Zhizibopi Decoction.Methods Chromatographic separation was performed on a Phenomenex Kinetex C_(18)column(150 mm×4.6 mm,2.6μm)with the mobile phase of acetonitrile-0.05 mol·L^(-1)potassium phosphate monobasic solution(pH adjusted to 3.0 by phosphoric acid),gradient elution was performed at a flow rate of 1.0 mL·min^(-1).The column temperature was 40℃.The detection wavelengths were set at 240 nm(for detection of genipin gentiobioside,geniposide and phellodendrine chloride),280 nm(for detection of 4-O-feruloylquinic acid,5-O-feruloylquinic acid,liquiritin and berberine hydrochloride)and 250 nm(for detection of glycyrrhizic acid).Results The mass concentrations of genipin gentiobioside,geniposide,phellodendrine chloride,4-O-feruloylquinic acid,5-O-feruloylquinic acid,liquiritin,berberine hydrochloride and glycyrrhizic acid showed good linear relationships with the peak area at 15.57-185.9,59.18-706.6,5.316-63.48,14.10-168.3,11.92-142.4,3.078-36.76,20.57-245.6 and 6.150-73.43 mg·L^(-1),respectively(r>0.999).The average recoveries of eight components were 102.1%,99.7%,98.3%,94.8%,99.5%,99.6%,98.2%and 103.5%,respectively,and the RSDs were all lower than 2%(n=6).Conclusion The HPLC wavelength switching method established in this study is simple and feasible,which can be used for simultaneous determination of the above 8 components in Zhizibopi Decoction.
