摘要
为了诊断西藏自治区日喀则市某牧场养殖藏羊的死亡病因,本试验无菌采集病死羊的心脏、肝脏、肺脏和肾脏病变组织进行细菌分离纯化,染色镜检和生化鉴定,16S rDNA序列分析,并对分离菌株进行药敏试验。结果显示,分离菌株革兰染色镜检呈葡萄串状或单个短链成对排列,可分解糖类,MR-VP结果为阳性,16S rDNA测序结果与GenBank上登录的其他金黄色葡萄球菌16S rDNA基因序列的同源性达98%以上,证明所分离菌株为金黄色葡萄球菌;药敏试验结果显示,分离菌株对部分喹诺酮类和头孢类抗生素、庆大霉素、复方新诺明、万古霉素和氯霉素高度敏感,对头孢他啶和多西环素呈中介耐药,而对氨苄西林、多黏菌素B、四环素、新霉素、卡那霉素、青霉素、克林霉素和红霉素共8种抗菌药呈耐药。本试验为日喀则地区羊源金黄色葡萄球菌的用药和治疗提供了参考依据。
In order to diagnose the cause of death in sheep from a farm in Shigatse city,Tibet Autonomous Region,this study collected sterile samples of heart,liver,lung,and kidney tissues from dead sheep for bacterial isolation and purification.The isolated bacteria were subjected to staining,microscopic examination,biochemical identification,16S rDNA sequencing,and drug sensitivity test.The results showed that the isolated strain exhibited Gram-positive staining,forming grape-like clusters or short chains in pairs.It was capable of sugar fermentation,and the MR-VP test was positive.Sequencing of the 16S rDNA showed over 98%homology with other registered 16S rRNA sequences of Staphylococcus aureus in GenBank,confirming the isolate as Staphylococcus aureus.The drug sensitivity test revealed that the isolated strain was highly sensitive to certain quinolones,cephalosporins,gentamicin,compound sulfamethoxazole,vancomycin and chloramphenicol,intermediate resistance to ceftazidime and doxycycline,while resistance to ampicillin,polymyxin B,tetracycline,neomycin,kanamycin,penicillin,clindamycin,and erythromycin.This study provides a reference for the medication and treatment of Staphylococcus aureus from sheep in the Shigatse region.
