摘要
研究旨在更好地合理科学化保护南疆沙蜥。试验利用微卫星标记手段对塔里木盆地5个地区的南疆沙蜥进行遗传多样性及遗传结构分析。结果显示,5个南疆沙蜥群体在4个微卫星位点观测杂合度、期望杂合度均值为0.464、0.692;多态信息含量均值为0.788;近交系数及群体分化系数均值分别为0.433、0.180。基于DAPC聚类分析表明,采自民丰县的南疆沙蜥群体单独聚类为一支,与聚类树的结果吻合。研究表明,5个地区的南疆沙蜥群体遗传多样性水平低下,群体间存在较大遗传分化。但由于样本量较少及采集区域不够丰富,有待进一步研究。试验预期成果可为保护南疆沙蜥遗传资源提供参考,也为其他荒漠化地区的物种种质资源保护奠定基础。
The aim of the study is to protect Phrynocephalus forsythii rationally and scientifically.The genetic diversity and genetic structure of Phrynocephalus forsythii from five regions of Tarim Basin were analyzed by using microsatellite markers.The results showed that the average observed heterozygosity and expected heterozygosity of five populations were 0.464 and 0.692 at four microsatellite loci.The mean polymorphic information content was 0.788.The mean inbreeding coefficient and population differentiation coefficient were 0.433 and 0.180,respectively.The DAPC cluster analysis showed that the population of Phrynocephalus forsythii from Minfeng County was clustered into a single branch,which was consistent with the results of the cluster tree.The study indicates that the genetic diversity of the population in the five regions is low,and there is a large genetic differentiation among the populations.However,due to the small sample size and insufficient collection area,further research is needed.The expected results of the experiment can provide a reference for the protection of genetic resources of Phrynocephalus forsythii and lay a foundation for the protection of species germplasm resources in other desertification areas.
