白花蛇舌草降低胃癌细胞化疗耐药的机制 被引量:2

摘要 目的研究白花蛇舌草对胃癌细胞化疗耐药的影响及可能的机制。方法取对数期SGC-7901细胞,建立顺铂耐药模型,分为耐药组、白花蛇舌草组、激活剂组、联合组。耐药组不作处理,白花蛇舌草组加入白花蛇舌草注射液(50μg·mL^(-1)),激活剂组加入信号转导及转录激活子3(signal transducer and activator of transcrption 3,STAT3)/神经源性基因Notch同源蛋白1(neurogenic gene Notch homolog 1,Notch1)轴激活剂Jagged1(500 ng·mL^(-1)),联合组加入白花蛇舌草注射液(50μg·mL^(-1))及Jagged1(500 ng·mL^(-1))。检测各组细胞的增殖抑制情况、细胞凋亡率、细胞葡萄糖摄取率、细胞相对乳酸含量及细胞STAT3和Notch1蛋白表达水平。结果与耐药组比较,白花蛇舌草组细胞增殖抑制率、凋亡率升高,半数抑制浓度(IC_(50))、葡萄糖摄取率、相对乳酸含量、STAT3和Notch1蛋白表达水平降低;激活剂组细胞增殖抑制率、凋亡率降低,IC_(50)、葡萄糖摄取率、相对乳酸含量、STAT3和Notch1蛋白表达水平升高(P<0.05)。与白花蛇舌草组比较,联合组细胞增殖抑制率、凋亡率降低,IC_(50)、葡萄糖摄取率、相对乳酸含量、STAT3、Notch1蛋白表达水平升高(P<0.05);与激活剂组比较,联合组细胞增殖抑制率、凋亡率升高,其他4项数值均降低(P<0.05)。结论白花蛇舌草可提高顺铂对胃癌细胞的增殖抑制率、减弱糖酵解并促进其凋亡、降低胃癌细胞化疗耐药,其作用机制可能与抑制STAT3/Notch轴活性有关。 Objective To Study the effect of Hedyotis diffusa on chemotherapy resistance of gastric cancer cells and the possible mechanism.Methods SGC-7901 cells in logarithm phase were taken to establish a cisplatin resistance model,which were divided into resistance group,Hedyotis diffusa group,activator group,and combination group.The drug resistance group was left untreated,the Hedyotis diffusa group was added with Hedyotis diffusa Injection(50μg·mL^(-1)),and the activator group was ad-ded with signal transducer and activator of transcription 3(STAT3)/neurogenic gene Notch homolog 1(Notch 1)axis activator Jagged1(500 ng·mL^(-1)),the combination group was added Hedyotis diffusa Injection(50μg·mL^(-1))and Jagged1(500 ng·mL^(-1)).The proliferation inhibition,cell apoptosis rate,cell glucose uptake rate,cell relative lactate content,and cell STAT3 and Notch1 protein expressions were detected in each group.Results Compared with the drug resistace group,the cell proliferation inhibition rate and apoptosis rate of each concentration were increased,the IC_(50),glucose uptake rate,relative lactate content,STAT3,Notch1 protein expression were decreased in the Hedyotis diffusa group,and the cell proliferation inhibition rate and apoptosis rate of each concentration were decreased,the IC_(50),glucose uptake rate,relative lactate content,STAT3,Notch1 protein expression were increased in the activator group(P<0.05).Compared with the Hedyotis diffusa group,the cell proliferation inhibition rate and apoptosis rate of each concentration were decreased,and the IC_(50),glucose uptake rate,relative lactate content,STAT3,Notch1 protein expression were increased in the combination group(P<0.05).Compared with the activator group,the cell proliferation inhibition rate and apoptosis rate of each concentration were increased,the other 4 values were decreased in the combination group(P<0.05).Conclusion Hedyotis diffusa can increase the proliferation inhibition rate of cisplatin on gastric cancer cells,reduce glycolysis and promote their apo
