摘要
试验旨在通过非标记定量技术对非澳毛皮海狮、南美毛皮海狮、智利毛皮海狮的胡须进行蛋白组学分析,探讨3种海狮间的亲缘关系。首先采集海狮胡须样品,提取总蛋白,再对蛋白进行浓度测定、电泳、酶解和肽段除盐等处理。得到的样品进行色谱与质谱分析、数据库检索、相关性分析与层次聚类分析、差异蛋白筛选与GO富集分析。结果表明:南美毛皮海狮N1与智利毛皮海狮Z1蛋白差异不明显、相关性强,自身表达量分布均匀;非澳毛皮海狮F1与这两种毛皮海狮间蛋白差异较为显著、相关性弱,自身表达量相对分布不均匀。说明非澳毛皮海狮和其他两种海狮亲缘关系较远,有相对明显的遗传差异,属于海狮科海狗属;而南美毛皮海狮与智利毛皮海狮的亲缘关系较近,可能为不同亚种。
The aim of this study was to analyze the proteomic whiskers of Non-Arctocephalus pusillus,Arctocephalus australis and Arctocephalus philippi by label-free quantitative technology,and to explore the relationship between the three species of fur seal.The total protein was extracted from the beard samples of fur seal,and then the protein concentration was determined,electrophoresis,enzymolysis and peptide desalting were performed.Then the samples were analyzed by chromatography and mass spectrometry,database search,correlation analysis and hierarchical cluster analysis,differential protein screening and GO enrichment analysis.The results suggested that no obvious expression difference in proteins of beards between Arctocephalus australis and Arctocephalus philippi was found,showing a strong correlation and uniform self-expression.However,Non-Arctocephalus pusillus was found significantly different from Arctocephalus australis and Arctocephalus philippi,showing a weak correlation and an uneven self-expression.It could be deduced that the Non-Arctocephalus pusillus could be a different species under the seal family,showing relatively significant genetic differences and a distant genetic relationship with Arctocephalus australis and Arctocephalus philippi,which were found having a closer genetic relationship and could be classified a different subspecies.
