三七皂苷对动脉损伤后血管内膜炎症、应激和增生的影响

Effects of Panax notoginseng saponins on vascular intimal inflammation,stress,and hyperplasia afterarterial injury

摘要 目的探讨三七皂苷对经皮动脉腔内血管成形术后炎症、应激和增生的影响。方法 30只新西兰兔颈动脉模型, 10只作为对照组, 20只大白兔用球囊制备颈动脉损伤模型, 随机数字表法分为模型组和三七皂苷组, 每组10只。三七皂苷组给予腹腔注射三七皂苷R1 50 mg/kg, 对照组和模型组给予等体积生理盐水。治疗4周后, 采用酶联免疫吸附试验(ELISA)试剂盒测定3组炎性因子肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β和IL-6的水平;采用放免法测定3组氧化应激指标超氧化物歧化酶(SOD)、谷胱甘肽氧化物酶(GSH-px)和丙二醛(MDA)的水平。HE染色分析血管增生情况。蛋白质免疫印迹分析血管核因子-κB(NF-κB)信号通路变化。计量数据比较采用t检验。结果三七皂苷组血管内膜面积[(0.15±0.02) mm2]明显低于对照组[(0.30±0.03) mm^(2)], 差异有统计学意义(t=14.300, P<0.05)。三七皂苷组血管内膜/中膜比(1.46±0.17)明显低于模型组(2.69±0.21), 差异有统计学意义(t=14.240, P<0.05)。三七皂苷组大白兔血清中IL-1β、IL-6和TNF-α水平[(78.36±13.70)、(67.80±12.44)、(47.40±13.34) ng/ml]明显低于模型组[(129.50±15.17)、(133.40±14.79)、(73.40±13.34) ng/ml], 差异有统计学意义(t=7.676、10.320、3.919, P<0.05)。三七皂苷组大白兔血清中SOD和GSH-Px水平[(36.10±5.02)、(45.90±7.06) ng/ml]明显高于模型组[(24.70±5.33)、(25.90±5.15) ng/ml], 差异有统计学意义(t=25.820、7.224, P<0.05)。三七皂苷组大白兔血清中MDA水平[(4.68±0.74) nmol/ml]明显低于模型组[(7.51±0.53) nmol/ml], 差异有统计学意义(t=9.854, P<0.05)。三七皂苷组大白兔血管磷酸化p65和p50水平(1.45±0.13、0.91±0.07)明显低于模型组(2.01±0.13、1.57±0.18), 差异有统计学意义(t=9.589、10.490, P<0.05)。结论三七皂苷可显著抑制动脉损伤后血管增殖、炎症和氧化应激, 主要通过NF-κB通路来实现的。 Objective To investigate the effects of Panax notoginseng saponins on vascular intimal inflammation,stress,and hyperplasia after arterial injury.Methods Of 30 New Zealand rabbits,10 served as control group,and 20 were used to prepare carotid artery injury models using balloons.They were divided into a model group and a Panax notoginseng saponin group by random number table.The rab-bits in the panax notoginseng saponin group were given intraperitoneal injection of Panax notoginseng sapo-nin(50 mg/kg),and those in the control and model groups were given an equal volume of physiological sa-line.After 4 weeks of treatment,the inflammatory factors tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and interleukin-6(IL-6)were measured by enzyme linked immunosorbent assay(ELISA).The levels of superoxide dismutase(SOD),glutathione oxidase(GSH px)and malondialdehyde(MDA)were measured by radioimmunoassay.The vascular proliferation was analyzed by HE staining.The nuclear factor-kB(NF-kB)signaling pathway was analyzed by Western blotting.The measurement data were com-paredbyt-test.Results The intimal area of blood vessels in the Panax notoginseng saponin group[(0.15±0.02)mm^(2)]was significantly smaller than that in the control group[(0.30±0.03)mm^(2),t=14.300,P<0.05].The ratio of vascular intima to media in the Panax notoginseng saponin group(1.46±0.17)was significantly lower than that in the model group(2.69±0.21,t=14.240,P<0.05).The levels of IL-1β,IL-6 and TNF-αin serum of white rabbits treated with Panax notoginseng saponins[(78.36±13.70),(67.80±12.44),(47.40±13.34)ng/ml]were significantly lower than those of the model group[(129.50±15.17),(133.40±14.79),(73.40±13.34)ng/ml,t=7.676,10.320,3.919,P<O.05].The levels of SOD and GSH-Px in the serum of white rabbits in the Panax notoginseng saponin group[(36.10±5.02),(45.90±7.06)ng/ml]were significantly higher than those in the model group[(24.70±5.33),(25.90±5.15)ng/ml,t=25.820,7.224,P<0.05].The MDA level in the serum of white rabbits in the Panax notogin
