脂肪间充质干细胞对乳腺癌癌前病变大鼠乳房微循环及血液流变学的影响

摘要 目的:观察脂肪间充质干细胞对乳腺癌前病变大鼠乳房微循环及血流动力学的影响,探讨脂肪间充质干细胞移植隆乳的安全性。方法:选择108只大鼠随机均分为健康组(n=36)、实验组(n=36)、对照组(n=36),其中实验组与对照组建成乳腺癌前病变模型,成模第10周对实验组大鼠第5对乳腺组织内注射脂肪间充质干细胞1×106个,对照组相同位置注射0.9%氯化钠溶液0.1 mL。第14周对三组大鼠进行血液流变学及血流动力学分析。结果:实验组大鼠中切全血黏度、低切全血黏度、血浆黏度、低切还原黏度低于对照组(P<0.05),与健康组比较无显著差异(P>0.05);微循环灌注量高于对照组(P<0.05),与健康组比较无显著差异(P>0.05)。结论:脂肪间充质干细胞移植对腺癌癌前病变模型大鼠的乳房微循环灌注量、血液流变学均有明显的改善作用。 Objective To observe the effects of adipose-derived mesenchymal stem cells on breast microcirculation and hemodynamics in rats with breast cancer precancerous lesions,and to explore the safety of breast augmentation with adipose-derived mesenchymal stem cells transplantation.Methods A total of 108 rats were randomly divided into a healthy group,an experimental group and a control group.72 rats in the experimental group and the control group were used to establish a model of breast cancer precancerous lesions.At the 10th week of modeling,the fifth pair of breast tissues of the experimental group rats were injected with adipose tissue derived mesenchymal stem cell 1×106,the control group was injected with 0.1 mL of physiological saline(0.9%wt/vol)at the same location.At the 14th week,hemorheology and hemodynamics of the three groups of rats were analyzed.Results The mid cut whole blood viscosity,low cut whole blood viscosity,plasma viscosity,and reduced viscosity of the experimental group rats were lower than those of the control group(P<0.05),and there was no significant difference compared to those in the healthy group(P>0.05).The microcirculation perfusion volume was higher than that of the control group(P<0.05),and there was no significant difference compared to that of the healthy group(P>0.05).Conclusions Transplantation of adipose-derived mesenchymal stem cells can significantly improve the perfusion of breast microcirculation and hemorheology in rats with precancerous lesions of adenocarcinoma.
