催化合成二氟烷基二氢苯并呋喃的研究进展

摘要 二氢苯并呋喃结构骨架广泛存在于具有药用价值和生物活性分子的天然产物中,因而引起了合成化学家和药物化学家们的广泛关注。另外,二氟烷基因其独特化学和结构性质在改变药物活性方面具有重要的作用。基于二氟烷基和二氢苯并呋喃的重要性,通过一步反应高效地合成二氟烷基二氢苯并呋喃成为目前有机合成方法学研究的热点之一。关注近几年的发展,综述了过渡金属催化,光协同过渡金属催化和叔胺参与的非共价作用促进烯炔串联环化合成二氟烷基二氢苯并呋喃的研究进展,并对此类反应中的底物范围和机理进行剖析。为了启发更加高效和绿色环保的催化体系出现,最后总结展望了合成二氟烷基二氢苯并呋喃的发展趋势。 The skeleton of dihydrobenzofuran widely exists in natural products with medicinal value and bioactive molecules,which has attracted extensive attention of synthetic chemists and medicinal chemists.In addition,difluoroalkyl groups play an important role in modifying drug activity due to their unique chemical and structural properties.Based on the importance of difluoroalkyl and dihydrobenzofuran,the efficient synthesis of difluoroalkyl dihydrobenzofuran through one-step reaction has become one of the hot topics in the research of organic synthesis methodology.Focusing on the development in recent years,the research progress of transition metal catalysis,photocooperative transition metal catalysis and non-covalent interaction with tertiary amine to promote the series cyclization of alkynes to synthesize difluoroalkyl dihydrobenzofuran was reviewed,and the substrate scope and mechanism of substrates in such reactions were analyzed.In order to inspire the emergence of more efficient and environmentally friendly catalytic systems,the development trend of synthesis of difluoroalkyl dihydrobenzofuran was summarized and prospected.
