摘要
【目的】为了阐明吲哚丁酸(IBA)诱导青榨槭不定根发生的激素调控机制。【方法】依据青榨槭扦插生根过程的解剖学观察,分别于对照期和4个生根关键时期采集插穗生根部位树皮组织进行IBA处理下的比较转录组学研究和生理指标检测。【结果】4个对比组分别鉴定到6774、6256、4522、4571个差异基因。GO(Gene ontoloy)富集分析发现,碳水化合物代谢和不定根发育等条目在不定根表达和伸长阶段高度富集。KEGG(Kyoto encyclopedia of genes and genomes)富集分析显示参与玉米素、油菜素内酯生物合成以及植物激素信号转导途径的差异基因发挥着重要作用。激素含量检测结果表明,高浓度的生长素参与了不定根的诱导过程,而相对低水平的生长素有利于其后期发育;细胞分裂素和玉米素均在愈伤组织形成期出现转运峰;油菜素内酯在生根初期被显著诱导。【结论】IBA刺激青榨槭不定根启动与增强生长素运输相关,不定根形成早期需要一定水平的细胞分裂素积累才能促进生根,玉米素和油菜素内酯的生物合成促进了不定根原基的诱导和启动。
[Objective]In order to clarify the hormonal regulation mechanism of Indole-3-butyric acid(IBA)-induced adventitious root formation of Acer davidii.[Methods]Based on the anatomical observation of the cutting rooting process of Acer davidi,the bark tissues of cuttings were collected at the control period and four critical rooting periods for comparative transcriptome study and physiological index detection under IBA treatment.[Results]The results showed that 6774,6256,4522 and 4571 differentially expressed genes were identified in the four comparison groups,respectively.GO enrichment analysis showed that carbohydrate metabolism and adventitious root development were highly enriched in the expression and elongation stages of adventitious roots.KEGG enrichment analysis showed that differential genes involved in zeatin,brassinosteroid biosynthesis and plant hormone signal transduction pathways played an important role in the formation of adventitious roots.The results of endogenous hormone content detection showed that high auxin concentration was involved in the induction process of adventitious roots,while relatively low level of auxin was beneficial to its later development.The transport peaks of cytokinin and zeatin appeared during callus formation.Brassinosteroid was significantly induced at the early rooting stage.[Conclusion]IBA stimulated the adventitious roots initiation,which was related to the enhancement of auxin transport.In the early stage of adventitious roots formation,it was necessary to maintain a certain concentration of cytokinin to promote rooting.The biosynthesis of zeatin and brassinosteroid promotes the induction and initiation of adventitious root primordium.
