基于内源激素调控的木荷扦插生根研究 被引量:6

Study on Cutting Rooting Technology of Schima superba Based on Endogenous Hormone Regulation

摘要 以木荷Schima superba优树嫁接苗萌发嫩枝为插穗,分析了扦插生根过程中插穗解剖构造以及内源激素水平的变化。结果表明:普通木荷插穗(生根率≈40%)无潜伏根原基,扦插10天时诱导产生不定根原始体,但插穗表观形态无明显改变,而扦插20天时产生不定根原基、茎基部明显愈伤化膨大,扦插50天不定根分化完成。与难生根基因型(生根率<10%)相比,易生根基因型(生根率>80%)插穗中的IAA水平明显偏高,而ZR、GA、ABA无明显差异;扦插20天,插穗IAA较高,GA偏低;扦插50天,IAA、ABA较高,而ZR、GA较低。在0~100 mg/L GA合成抑制剂多效唑作用下,普通插穗生根率差异显著。其中,以50 mg/L多效唑处理下生根率最高,达90.1%,较对照提升了123.6%。研究认为,木荷属生根困难树种,内源性IAA、GA水平在不定根形成中起到关键作用,通过外源喷施多效唑能有效提高木荷生根能力,但鉴于GA在生根过程中的双重性作用,其施用浓度需控制在合适范围。 The changes in anatomical structure and endogenous hormone level of cuttings in the process of rooting were analyzed by using the sprouted twigs of grafted seedlings of Schima superba as cuttings,to provide a theoretical reference for the construction of an efficient rooting technology system of S.superba.The results showed that there was no latent root primordium in general cuttings(rooting rate≈40%)of S.superba.Adventitious root primordia were induced at 10 days of cutting,but there was no significant change in the apparent morphology of cuttings.Adventitious root primordia and stem base were significantly callused and expanded at 20 days of cutting,and adventitious root differentiation was completed at 50 days of cutting.Compared with the difficult rooting genotype(rooting rate<10%),the IAA level in the cuttings of the easy rooting genotype(rooting rate>80%)was significantly higher,but there was no significant difference in ZR,GA,and ABA.Cutting 20 days,cuttings IAA higher,GA lower;Cutting 50 days,IAA,ABA higher,while ZR,GA lower.Under the effect of 0-100 mg/L GA synthesis inhibitor,the rooting rate of general cuttings was significantly different.Among them,the rooting rate was the highest under the treatment of 50 mg/L paclobutrazol,reaching 90.1%,which increased by 123.6%compared with the control.The results showed that the endogenous IAA and GA levels played a key role in the formation of adventitious roots in S.superba,which was difficult to root.Spraying exogenous paclobutrazol could effectively improve the rooting ability of S.superba,but because of the dual role of GA in the rooting process,its application concentration should be controlled in the appropriate range.
