突发性聋患者血清干扰素调节因子9和沉默信息调节因子1水平及其临床意义

Serum levels of interferon regulatory factor 9 and silent information regulator 1 in patients with sudden sensorineural hearing loss and their clinical significance

摘要 目的分析突发性聋患者血清干扰素调节因子9(IRF9)和沉默信息调节因子1(SIRT1)的表达情况,并探究其临床意义。方法选取2020年3月至2022年3月湖北省中医院收治的150例突发性聋患者为病例组,并同期选取150名检查健康者为对照组。分析两组对象的一般资料;采用PCR法检测两组IRF9表达水平,酶联免疫吸附法(ELISA)检测SIRT1水平;采用Pearson分析突发性聋患者血清IRF9与SIRT1的相关性;采用ROC曲线分析IRF9、SIRT1水平对突发性聋患者的诊断价值;采用logistic回归分析影响突发性聋患者的因素。结果病例组饮酒人数多于对照组,收缩压(SBP)、舒张压(DBP)高于对照组,差异有统计学意义(P<0.05)。病例组的IRF9水平高于对照组,SIRT1水平低于对照组,差异有统计学意义(P<0.05)。相关性分析显示,突发性聋患者血清IRF9和SIRT1呈负相关(r=-0.576,P<0.05)。ROC分析显示,血清IRF9和SIRT1的ROC曲线的下面积(AUC)分别为0.738(95%CI=0.681~0.796)、0.746(95%CI=0.691~0.800)二者联合检测的AUC为0.818(95%CI=0.769~0.866),高于IRF9、SIRT1单独检测。多因素回归分析显示,IRF9是影响突发性聋患者的危险因素(OR=1.604,95%CI=1.221~2.106,P<0.001),SIRT1为保护因素(OR=0.729,95%CI=0.593~0.896,P=0.003)。结论突发性聋患者血清IRF9水平升高,SIRT1水平降低,二者水平对患者的早期临床诊断有重要意义。 Objective To analyze the expression of serum interferon regulatory factor 9(IRF9)and silent information regu-lator 1(SIRT1)in patients with sudden deafness,and to explore their clinical significance.Methods A total of 150 patients with sudden deafness admitted to Hubei Provincial Hospital of TCM from March 2020 to March 2022 were selected as the case group,and 150 healthy people were selected as the control group during the same period.The general data of the two groups were analyzed.The expression of IRF9 was detected by PCR,and the level of SIRT1 was detected by enzyme-linked immunosorbent assay(ELISA).Pearson was used to analyze the correlation between serum IRF9 and SIRT1 in patients with sudden deafness.Receiver operating characteristic(ROC)curve was used to analyze the diagnostic value of IRF9 and SIRT1 levels in patients with sudden deafness.Logistic regression was used to analyze the factors affecting patients with sudden deafness.Results The number of drinking in the case group was more than that in the control group,and the systolic blood pressure(SBP)and diastolic blood pressure(DBP)were higher than those in the control group,and the differences were sta-tistically significant(P<0.05).The level of IRF9 in the case group was higher than that in the control group,and the level of SIRT1 was lower than that in the control group,and the differences were statistically significant(P<0.05).Correlation anal-ysis showed that serum IRF9 was negatively correlated with SIRT1 in patients with sudden deafness(r=-0.576,P<0.05).ROC analysis showed that,the area under the ROC curve(AUC)of serum IRF9 and SIRT1 was 0.738(95%CI=0.681-0.796)and 0.746(95%CI=0.691-0.800),re-spectively.The AUC of combined detection of the two was 0.818(95%CI=0.769-0.866),and it was higher than IRF9 and SIRT1 alone.Multivariate regression analysis showed that IRF9 was a risk factor for sudden deafness(OR=1.604,95%CI=1.221-2.106,P<0.001),and SIRT1 was a protective factor(OR=0.729,95%CI=0.593-0.896,P=0.003).Conclusion Serum IRF9 level is increase
