摘要
目的建立柱前衍生蚕蛾中丙氨酸含量的测定方法,为不同产地蚕蛾品质评价和质量控制实验依据。方法采用柱前衍生高效液相色谱法。色谱柱为Diamonsil C18色谱柱(4.6 mm×200 mm,5μm);柱温20℃;流动相A为乙腈-水(1∶1),流动相B为0.05 mol/L醋酸钠缓冲液(pH=6.4),梯度;流速1.0 ml/min;检测波长为360 nm。结果丙氨酸进样量在0.04~0.2μg范围内呈良好线性,回归方程为Y=274444X+9597.5;相关系数r=0.9996。结论建立的含量测定方法专属性强、准确度高,重复性良好,可用于蚕蛾药材的质量控制。
Objective To establish a method for the determination of alanine in silkworm moth derived from precolumn,so as to provide experimental basis for quality evaluation and quality control of silkworm moth from different habitats.Methods High performance liquid chromatography with precolumn derivatization was used.The chromatographic column is Diamonsil C18 chromatographic column(4.6 mm×200 mm,5μm),column temperature:20℃.Mobile phase A is acetonitrile water(1∶1),mobile phase B is 0.05 mol/L sodium acetate buffer(pH=6.4),gradient;The flow rate is 1.0 ml/min.The detection wavelength is 360 nm.Results The alanine injection volume showed good linearity in the range of 0.04~0.2μg,and the regression equation was Y=274444X+9597.5.Correlation coefficient r=0.9996.Conclusion The method is specific,accurate and reproducible,and can be used for the quality control of silkworm moth.
