摘要
目的建立气相色谱法同时测定雌性柞蚕蛾中棕榈酸、棕榈油酸、硬脂酸、油酸、亚油酸、α-亚麻酸6种成分的含量。方法样品甲酯化后,采用Agilent VF-23 ms毛细管柱(30 m×0.25 mm,0.25μm),柱温程序升温,进样口:250℃,FID检测器:300℃,流速:0.6 m L·min^(-1),进样量:1μL。结果棕榈酸、棕榈油酸、硬脂酸、油酸、亚油酸、α-亚麻酸质量浓度分别在0.032~0.288、0.0064~0.058、0.0064~0.058、0.049~0.443、0.015~0.137、0.083~0.745 g·L^(-1)内呈良好的线性关系(r≥0.999 1),平均回收率为97.8%~99.6%(RSD≤1.7%,n=9)。结论该方法可用于控制雌性柞蚕蛾药材和饮片的质量。
Objective To develop a GC method for simultaneous analysis of six aliphatic acid components including palmitic acid, palmitoleic acid, stearic acid, oleic acid, linoleic acid and a-linolenic acid in Antheraea pernyi Female Moth. Methods The GC separation was achieved on a Agilent VF-23ms (30 m × 0. 25 mm ×0. 25 μm) column with a temperature program at the rate of 0.6 mL. min - 1. Flame-ionization detector temperature was 300 ℃ and injection port temperature was 250 ℃. Sample size was 1 μL. Results The linear range of palmitic acid, palmitoleic acid, stearic acid, oleic acid, linoleic acid and a-linolenic acid was 0. 032 -0. 288,0. 0064 -0. 058,0. 0064 -0. 058,0. 049 -0. 443,0. 015 -0. 137,and 0. 083 -0. 745 g.L-1 , respectively( r≥0. 999 1 ). The average recovery( n =9) was between 97.8% -99. 6% with RSD less than 1.7%. Conclusions The method is simple, accurate, reproducible, and could be used for quality control of Antheraea pernyi Female Moth.
