摘要
目的探讨喉鳞状细胞癌(LSCC)组织中tat结合蛋白30(TIP30)、dickkopf相关蛋白1(DKK1)的表达及其与病理特征和预后的关系。方法选取103例LSCC患者,采用实时荧光定量聚合酶链式反应检测LSCC组织和癌旁组织中TIP30 mRNA、DKK1 mRNA表达,分析二者与病理特征的关系。根据LSCC组织中TIP30 mRNA、DKK1mRNA表达均值分为高、低表达者,随访3年,采用Kaplan-Meier法绘制不同TIP30 mRNA、DKK1 mRNA表达患者生存曲线,采用多因素Cox回归分析LSCC患者死亡的影响因素。结果与癌旁组织比较,LSCC组织中TIP30 mRNA表达降低,DKK1 mRNA表达升高(P均<0.05)。不同组织分化程度、TNM分期、淋巴结转移LSCC组织中TIP30mRNA、DKK1 mRNA表达比较,差异有统计学意义(P均<0.05)。103例LSCC患者累积生存率为69.90%(72/103),K-M生存曲线显示,高TIP30 mRNA表达者3年累积生存率高于低表达者,高DKK1 mRNA表达者3年累积生存率低于低表达者(P均<0.05)。多因素Cox回归分析显示,中低分化、TNM分期Ⅲ期、淋巴结转移、DKK1 mRNA升高为LSCC患者死亡的独立危险因素,TIP30 mRNA升高为独立保护因素(P均<0.05)。结论LSCC组织中TIP30 mRNA低表达,DKK1 mRNA高表达,二者表达与组织分化程度、TNM分期、淋巴结转移和预后有关。
Objective To investigate the expression of Tat-interactive protein 30(TIP30)and dickkopf-related protein 1(DKK1)in laryngeal squamous cell carcinoma(LSCC)tissues and their relationships with clinicopathologic features and prognosis.Methods Totally 103 LSCC patients were selected.The expression levels of TIP30 mRNA and DKK1 mRNA in LSCC tissues and paraneoplastic tissues were detected by real-time fluorescence quantitative polymerase chain reaction,and the relationships between the two and pathological parameters were analyzed.According to the mean values of TIP30 mRNA and DKK1 mRNA expression in LSCC tissues,they were divided into the high and low expression groups.After 3 years of follow-up,the K-M method was used to plot the survival curves of patients with different TIP30 mRNA and DKK1 mRNA expression,and multi-factor Cox regression was used to analyze the factors influencing the death of LSCC patients.Results Compared with the paraneoplastic tissues,TIP30 mRNA expression decreased and DKK1 mRNA expression increased in the LSCC tissues(both P<0.05).There were significant differences in the expression levels of TIP30 mRNA and DKK1 mRNA between LSCC tissues with different differentiation degree,TNM stage and lymph node metastasis(all P<0.05).The cumulative survival rate of 103 patients with LSCC was 69.90%(72/103).The K-M survival curve showed that 3-year cumulative survival rate in the high TIP30 mRNA expression group was higher than that in the low expression group,and 3-year cumulative survival rate in the high DKK1 mRNA expression group was lower than that in the low expression group(both P<0.05).Multi-factor Cox regression analysis showed that moderate and low differentiation,TNM stage III,lymph node metastasis,and DKK1 mRNA were independent risk factors for death in LSCC patients,and TIP30 mRNA was an independent protective factor(all P<0.05).Conclusion The low expression of TIP30 mRNA and the high expression of DKK1 mRNA in LSCC tissues are related to the degree of differentiation,TNM stage,lymph node met
