摘要
目的探讨钙电压门控通道辅助亚基α2δ1作为喉鳞状细胞癌(laryngeal squamous cell carcinoma,LSCC)预后评价指标的可能性。方法选取2017年1月~2020年12月在沧州市人民医院接受诊治的LSCC患者80例作为研究对象。应用蛋白质印迹技术、免疫组织化学技术和免疫荧光染色技术检测喉癌组织、癌旁组织和正常喉黏膜组织样本中α2δ1蛋白的表达量。结果α2δ1蛋白在LSCC中高表达,在癌旁组织和正常喉黏膜组织中低表达(P<0.001)。LSCC患者癌组织中α2δ1蛋白表达在性别(P=0.559)、年龄(P=0.950)和是否存在吸烟史(P=0.314)的比较,差异无统计学意义。病理学分级中低分化肿瘤的α2δ1蛋白表达明显高于中高分化(P<0.05);T3、T4肿瘤组织中α2δ1蛋白值显著高于T1、T2肿瘤组织(P=0.005);存在淋巴结转移的病理组织中,α2δ1蛋白表达高于未发生淋巴结转移的组织(P<0.05);临床分期越高的患者,其肿瘤组织中α2δ1蛋白表达越高(P=0.0001)。患者生存分析曲线显示α2δ1高表达组患者预后较低表达组患者预后差(P=0.01)。结论α2δ1蛋白在LSCC中高表达,且与患者生存密切相关,可作为患者预后预测指标。
OBJECTIVE To explore the possibility of calcium voltage-gated channel auxiliary subunit(α2δ1)as a prognostic evaluation indicator of laryngeal squamous cell carcinoma(LSCC).METHODS Eighty patients with laryngeal squamous cell carcinoma diagnosed and treated in Cangzhou's Hospital from January 2017 to December 2020 were selected as the study subjects.The expression ofα2δ1 protein was detected by Western blot,immunohistochemistry and immunofluorescence staining assays.RESULTSα2δ1 protein was highly expressed in LSCC,and low expressed in paracancerous tissues and normal tissues(P<0.001).The expression ofα2δ1 protein in cancer tissues of patients with sex(P=0.559),age(P=0.950)and smoking history(P=0.314)was not statistically significant.The expression ofα2δ1 protein in poorly differentiated tumors was significantly higher than that in moderately differentiated tumors(P<0.05).The expression ofα2δ1 protein in T3/T4 tumor tissues was significantly higher than that in T1/T2 tumor tissues(P=0.005).The expression ofα2δ1 protein in the tissues with lymph node metastasis was higher than that in the tissues without lymph node metastasis(P<0.05).The higher the clinical stage,the higher the expression ofα2δ1 protein in tumor tissue(P=0.0001).Survival analysis showed that patients with highα2δ1 expression had a worse prognosis than those with lowα2δ1 expression(P=0.01).CONCLUSIONα2δ1 protein is highly expressed in LSCC and is closely related to patient survival,which can be used as a prognostic indicator.
