摘要
目的:建立测定中药茯苓4个药用部位(茯苓皮、赤茯苓、白茯苓和茯神)中多个三萜酸成分(茯苓酸、茯苓新酸B、去氢土莫酸、茯苓新酸A、去氢茯苓酸和松苓新酸)的一测多评(quantitative analysis of multi-components with a single-marker,QAMS)方法,验证该方法在茯苓质量分析中的可行性与科学性。方法:在建立的变波长-梯度洗脱HPLC方法的基础上,以茯苓酸为内参物,建立内参物与茯苓新酸B、去氢土莫酸、茯苓新酸A、去氢茯苓酸和松苓新酸的相对校正因子(f_(i/s)),并采用外标法(external standard method,ESM)与QAMS法分别测定不同产地茯苓4个药用部位样品中6个三萜酸成分的含量。采用Origin 2021进行相似度分析和聚类分析。结果:方法学考察表明所建立的变波长-梯度洗脱HPLC方法准确可靠;各相对校正因子重复性良好,QAMS法与ESM测定结果之间无显著差异。相似度和聚类分析结果表明,茯苓皮样品的产地差异较大,其中云南产的茯苓皮相似度相对较好品质较均一,赤茯苓、白茯苓和茯神的产地差异较小;不同药用部位间6个茯苓三萜酸含量的差异较大,茯苓皮中三萜酸的含量最高,赤茯苓次之,其次为白茯苓和茯神;在同一药用部位的6个茯苓三萜酸含量方面,茯苓皮中松苓新酸和茯苓酸的含量相对较高,赤茯苓、白茯苓及茯神中以茯苓酸、去氢茯苓酸和去氢土莫酸占优势,赤茯苓中的茯苓新酸A含量相对最低。结论:建立的变波长-梯度洗脱HPLC分析策略和QAMS方法可以同时评价茯苓4个药用部位中6个茯苓三萜酸成分,对于中药茯苓中的三萜成分分析和质量控制具有重要参考价值。
Objective:To establish a new method for the quantitative analysis of multi-components by quantitative analysis of multi-components with a single-marker(QAMS)and to determine the main triterpenes(pachymic acid,poricoic acid B,dehydrotumulosic acid,poricoic acid A,dehydropachymic acid,dehydrotrametenolic acid)in four medicinal parts(Poriae Cutis,Rubra Poria,White Poria,Poria cum Radix Pini)of Poria cocos simultaneously,and verify the feasibility and scientificity of the method in the quality analysis of P.cocos.Methods:Based on the established variable wavelength-gradient elution HPLC method,pachymic acid was chosen as the reference for establishing the relative correction factor of poricoic acid B,dehydrotumulosic acid,poricoic acid A,dehydropachymic acid,dehydrotrametenolic acid.The contents of six components in four medicinal parts of P.cocos from different habitats were determined by both external standard method(ESM)and QAMS.Origin 2021 was used for similarity analysis and cluster analysis.Results:The established variable wavelength-gradient elution HPLC method was accurate and reliable,and the repeatability of each relative correction factor was excellent.There was no significant difference between the results calculated by QAMS method and ESM.The results of similarity and cluster analysis showed that there were great differences in the habitats of Poriae Cutis,among which,the similarity of yunnan Poriae Cutis was relatively high and the quality was relatively uniform,while the differences of the habitats of Rubra Poria,White Poria and Poria cum Radix Pini were relatively small.The content of six triterpenes varied greatly among different medicinal parts of P.cocos.The triterpenes content of Poriae Cutis was the highest,followed by Rubra Poria,White Poria and Poria cum Radix Pini.In the same medicinal part of P.cocos,the contents of six triterpenes were as follows.The contents of dehydrotrametenolic acid and pachymic acid in Poriae Cutis were relatively high,and the contents of pachymic acid,dehydropachymic a
