斜率校正一测多评法同时测定茯苓中3种三萜酸成分的含量 被引量:1

摘要 目的建立新的一测多评法同时测定茯苓中3-O-乙酰基-16α-羟基-氢化松苓酸、去氢茯苓酸和茯苓酸的含量。方法以Agilent ZORBAX SB-C_(18)(4.6 mm×250 mm,5µm)为色谱柱,以乙腈-0.1%磷酸水溶液为流动相等度洗脱;流速:0.8 mL/min;进样量:10µL;柱温:30℃;检测波长为210 nm。以茯苓酸为内参计算其他两个成分的校正因子。结果以茯苓酸为内参,3-O-乙酰基-16α-羟基-氢化松苓酸和去氢茯苓酸的校正因子分别为0.71和1.43;一测多评法测定结果与外标法测定结果无明显差异[相对误差(RE)<5%]。结论建立的一测多评方法简便准确,可用于茯苓药材中多指标成分的含量测定,为茯苓药材质量评价提供参考。 Objective To establish a new method for simultaneous determination of 3-O-acetyl-16α-hydroxytrametenolic acid,dehydropachymic acid and pachymic acid in Poria by quantitative analysis of multi-components with single marker(QAMS).Methods The determination was performed on Agilent ZORBAX SB-C_(18)(4.6 mm×250 mm,5µm)column with the mobile phase consisted of acetonitrile-0.1%phosphoric acid aqueous solution at the flow rate of 0.8 mL/min.The injection volume was 10µL.The column temperature was set at 30℃.The detection wavelength was set at 210 nm.The correction factors of the other two components were calculated by using pachymic acid as the internal reference.Results The correction factors of 3-O-acetyl-16α-hydroxytrametenolic acid and dehydropachymic acid calculated by the internal reference pachymic acid were 0.71 and 1.43,respectively.The measurement data of QAMS were not significantly different from those of the external standard method(relative error<5%).Conclusion The method is simple and accurate,and can be used for the determination of multi-components in Poria,providing a reference for the quality evaluation of Poria.
