摘要
目的探讨有氧运动和日粮限制对肥胖大鼠减重效果及对比目鱼肌MSTN的影响,为运动和日粮限制对肥胖机体减重及骨骼肌的影响提供理论依据。方法高脂喂养建立肥胖模型后分为安静肥胖组、有氧运动组和日粮限制组,记录大鼠体重、肌肉质量,HE染色观察比目鱼肌横截面积,试剂盒测定大鼠血清血脂4项、NEFA、MSTN、FST水平,WB测定比目鱼肌MSTN和FST蛋白表达。结果有氧运动和饮食限制均可有效降低肥胖大鼠的体重,有氧运动可显著增加比目鱼肌横截面积、血清HDL和FST水平、比目鱼肌FST蛋白表达;显著降低血清MSTN水平,比目鱼肌MSTN蛋白表达。日粮限制可显著降低肌肉质量、血清LDL和FST水平;显著增加比目鱼肌横截面积和质量指数。结论有氧运动和日粮限制均可显著降低肥胖大鼠的体重,有氧运动可通过增加FST降低MSTN促进比目鱼肌横截面积增加,日粮限制对MSTN无影响。
Objective To investigate the effect of aerobic exercise and diet restriction on weight loss and the effect on MSTN of flounder muscle in obese rats,and to provide a theoretical basis for the effect of exercise and diet restriction on weight loss and skeletal muscle in obese organisms.Methods After high-fat feeding to establish obesity model,rats were divided into quiet obesity group,aerobic exercise group and diet restriction group,and their body weight and muscle mass were recorded.The cross-sectional area of the flounder muscle was observed by HE staining,the levels of serum NEFA,MSTN,FST and Leptin were measured by the kit,and the expression of MSTN and FST in flounder muscle was measured by WB.Results Both aerobic exercise and dietary restriction were effective in reducing body weight in obese rats.Aerobic exercise significantly increased flounder muscle cross-sectional area,serum HDL and FST levels,and flounder muscle FST protein expression;significantly decreased serum MSTN levels,and flounder muscle MSTN protein expression.Dietary restriction significantly reduced muscle mass,serum LDL and FST levels;significantly increased flounder muscle cross-sectional area and mass index.Conclusion Aerobic exercise and diet restriction both significantly reduced body weight in obese rats,and aerobic exercise decreased MSTN by increasing FST to promote the increase of flounder muscle cross-sectional area,while diet restriction had no effect on MSTN.
