摘要
目的探讨帕金森病(pakinson disease,PD)患者粪便微生物菌落组成及变化。方法选取2019年4月—2020年7月期间就诊中国医科大学大学附属第一医院和沈阳市第四人民医院的帕金森患者(PD组)20例为研究对象,其20名配偶作为健康对照组(HC组),采用16S rRNA高通量测序技术对20例帕金森病患者及其健康配偶粪便中的微生物菌落进行分析。结果Alpha多样性分析中,PD组Chao1多样性指数(1114.556±315.308)、Observed species指数(959.885±294.012)、Shannon指数(6.357±0.928)、Simpson指数(0.928±0.063),与对照组相比,差异有统计学意义(t=2.949、2.666、3.419、2.491,P<0.05)。在门水平上,PD组放线菌门相对丰度及变形菌门相对丰度明显高于对照组,而PD组厚壁菌门相对丰度明显低于对照组,差异有统计学意义(P<0.05);而在属水平上,PD组中监测到18种显著升高的肠道菌群(P<0.05)。结论PD患者粪便微生物群生态失调,阐明粪便微生物菌落的差异将有助于提高我们对PD发病机制中微生物群的理解。
Objective To investigate the composition and changes of fecal microbial colonies in patients with parkinson disease(PD).Methods 20 PD patients admitted to the First Affiliated Hospital of China Medical University and Shenyang Fourth People´s Hospital from April 2019 to July 2020 were selected as the research objects,and 20 of their spouses were served as healthy controls group.The fecal microbial communities of 20 patients with Parkinson´s disease and their healthy spouses were analyzed by 16S rRNA high-throughput sequencing technology.Results In the Alpha diversity analysis,Chao1 diversity index of PD group(1114.556±315.308),Observed species index(959.885±294.012),Shannon index(6.357±0.928),Simpson index(0.928±0.063),compared with the control group,the difference was statistically significant(t=2.949,2.666,3.419,2.491,P<0.05).At the phylum level,the relative abundance of actinomycetes and Proteobacteria in PD group was significantly higher than that in control group,while the relative abundance of firmicutes in PD group was significantly lower than that in control group,the difference was statistically significant(P<0.05).At the genus level,18 species of intestinal flora were significantly increased in PD group(P<0.05).Conclusion The ecological imbalance of fecal microbiota in PD patients,and elucidating the differences of fecal microbial colonies will help to improve our understanding of the microbiota in the pathogenesis of PD.
