摘要
本研究采用农业行业标准《大麦品种鉴定技术规程SSR分子标记法》(NY/T 2466-2013)中的28个标记对142份大麦申请品种进行DNA指纹采集和遗传多样性分析。28个标记共检测到196个等位变异和314个基因型,平均值分别为7和11.2,PIC值范围为0.1406~0.7904,平均值为0.5853。聚类结果显示,142份材料可以完全区分,遗传距离范围为0.0314~0.9747,平均值为0.6172。本研究分别以字符串形式和条形码格式表示142份大麦品种的DNA指纹图谱,每一份品种的指纹都具有唯一性,指纹数据可作为大麦新品种保护受理审查的参考依据和技术支撑,有利于大麦育种创新和新品种推广。
Based on the industry standard"Identification of barley varieties-SSR method"(NY/T 2466-2013),we carried out DNA fingerprint construction and genetic diversity analysis on 142 barley varieties with 28 markers.Total of 196 alleles and 314 genotypes could be detected by using these 28 pairs of markers,with an average value of 7 and 11.2,respectively.PIC values ranged from 0.1406 to 0.7904,with a mean value of 0.5853.The range of polymorphism information content(PIC)at 28 markers were 0.1406~0.7904,with an average of 0.5853.Cluster analysis showed that,142 varieties could be completely identified,the range of genetic distance was 0.0314~0.9747,with an average value of 0.6172.According to the result,we constructed the fingerprint map of the 142 varieties in the form of string and barcode.The fingerprint of each variety is unique,which provided reference basis and technical support for barley variety protection.Our study would benefit barley breeding and promotion of new variety.
