摘要
目的研究miRNA-23b-3p在不明原因复发性流产(URSA)蜕膜组织中的表达并分析其可能的作用机制。方法选取2021年10月至2022年10在内蒙古自治区人民医院因URSA稽留流产行清宫术的患者及无生育计划行人工流产术的正常妊娠者各3例,均收集蜕膜组织。比较两组蜕膜组织中miRNA-23b-3p表达水平,利用生物信息学方法预测miRNA-23b-3p潜在靶基因和相关信号通路。结果URSA组miRNA-23b-3p的表达水平较正常妊娠人工流产患者明显升高(P<0.01);利用生物信息学方法分析发现,miRWalk、RNAInter和RNA223个数据库具有2359个miRNA-23b-3p的靶基因交集;通过GO及KEGG功能富集及信号通路分析,miRNA-23b-3p靶基因主要富集于Wnt信号通路(P<0.05),并且2359个交集靶基因中38个靶基因富集于Wnt信号通路。结论蜕膜组织中miRNA-23b-3p可能通过调控Wnt信号通路参与URSA发生发展,为后续URSA防治策略研究提供了新的思路和靶点。
ObjectiveTo investigate the expression of miRNA-23b-3p in decidual tissue of unexplained recurrent abortion(URSA)and analyze its possible mechanism.Methods From October 2021 to October 2022,decidual tissue was collected from 3 patients with missed abortion due to URSA who underwent curettage and 3 normal pregnancies who underwent artificial abortion without family planning.Compare the expression level of miRNA-23b-3p in decidual tissue of two groups,and predict the potential target genes and related signal pathways of miRNA-23b-3p using bioinformatics methods.Results The expression level of miRNA-23b-3p in URSA group was significantly higher than that in normal pregnancy induced abortion patients(P<0.01);Using bioinformatics methods,it was found that the miRWalk,RNACenter,and RNA22 databases had 2359 miRNA-23b-3p target gene intersections;Through functional enrichment and signal pathway analysis of GO and KEGG,miRNA-23b-3p target genes were mainly enriched in the Wnt signal pathway(P<0.05),and 38 of 2359 intersection target genes were enriched in the Wnt signal pathway.Conclusion miRNA-23b-3p in decidual tissue may participate in the occurrence and development of URSA by regulating the Wnt signaling pathway,providing a new idea and target for further research on prevention and treatment strategies for URSA.
