摘要
目的:观察强骨宝对去卵巢大鼠疲劳性损伤诱发膝骨关节炎(KOA)的作用,以探讨该方对关节软骨的保护机制。方法:将24只SD大鼠随机分为空白对照组、模型组及强骨宝组,每组8只。空白对照组不进行手术干预,模型组及强骨宝组均采用去卵巢联合跑台运动构建KOA模型。术后连续灌胃30 d,于第30天灌胃后行大鼠膝关节MRI影像学检测并取材。采用HE染色及Mankin评分观察、评价膝关节病理学改变,ELISA法检测血清中白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)及软骨寡聚基质蛋白(COMP)含量,Western blot法检测关节软骨中基质金属蛋白酶-3(MMP-3)及含Ⅰ型血小板结合蛋白基序的解聚蛋白样金属蛋白酶-7(ADAMTs-7)的表达情况。结果:与模型组比较,强骨宝组MRI关节液信号减弱,且无关节囊局部信号增强情况;强骨宝组软骨表面更加平滑,潮线层次清晰,炎症细胞浸润减少;关节软骨Mankin评分显著降低(P<0.01);血清中IL-1β、TNF-α、COMP含量均显著下降(P<0.01);关节软骨中MMP-3、ADAMTs-7蛋白表达显著降低(P<0.01)。结论:强骨宝可有效降低KOA大鼠IL-1β、TNF-α、MMP-3、ADAMTs-7的表达水平,通过抑制炎症反应,减少软骨中COMP的降解,从而达到保护关节软骨的作用。
Objective:The effect of Qianggubao on knee osteoarthritis(KOA)induced by fatigue injury in ovariectomized rats was observed to explore the protective mechanism of this prescription on articular cartilage.Methods:A total of 24 SD rats were randomly divided into blank control group,model group and Qianggubao group,8 rats in each group.The blank control group underwent no surgical intervention,and both the model group and the Qianggubao group used the ovariecolectomy method and the combined platform running exercise to construct the KOA model.It was performed for 30 d,and MRI imaging was performed on day 30.HE staining and Mankin score were adopted to observe and evaluate the pathological changes of knee joints.ELISA method was used to detect the contents of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),and cartilage oligomeric matrix protein(COMP)in serum.The Western blot method was used to detect the expression of matrix metalloproteinases-3(MMP-3)and a disintegrin-like and metalloproteinase with thrombospondin type1 motif-7(ADAMTs-7)in the cartilage.Results:Compared with the model group,the signal of joint fluid in the Qianggubao group was weaker,and there was no local signal enhancement of the joint capsule;the cartilage surface of the Qianggubao group was smoother,the tide line layers were clear,and the infiltration of inflammatory cells was reduced.The Mankin score of the Qianggubao group was significantly decreased(P<0.01);the serum levels of IL-1β,TNF-αand COMP were significantly decreased(P<0.01);the protein expression of MMP-3 and ADAMTs-7 in the cartilage were significantly decreased(P<0.01).Conclusion:Qianggubao can effectively reduce the expression levels of IL-1β,TNF-α,MMP-3 and ADAMTs-7 in KOA rats.By inhibiting the inflammatory response,and reducing the degradation of COMP in the cartilage,it can protect the articular cartilage.
