摘要
目的通过研究阿戈美拉汀(AGO)对抑郁模型小鼠体内过氧化氢酶(CAT)活性及其对氧化应激的影响,进一步分析阿戈美拉汀改善抑郁的具体机制。方法在4周内建立慢性束缚应激(CRS)模型小鼠,同时按照具体分组进行阿戈美拉汀50 mg/(kg·d),地拉罗司(DFX)10 mg/(kg·d)灌胃处理。4周后进行行为学测试。同时,使用Western Blot和免疫组化法评估CAT、氧化应激信号通路的表达情况。结果AGO治疗4周可明显改善小鼠的抑郁样行为,同时,激活CRS小鼠海马和血清中的CAT,缓解氧化应激损伤,而DFX可以部分抵消掉这种治疗作用。结论AGO可以通过改变CAT活性影响氧化应激从而发挥抗抑郁作用。
Objective To further analyze the specific mechanism of agomelatine in improving depression by studying the effect of agomelatine(AGO)on the activity of catalase(CAT)in the body of depression model mice and its effect on oxidative stress.Methods The model mice of chronic restraint stress(CRS)were established within 4 weeks.Agomelatine 50 mg/(kg·d)and desloratose(DFX)10 mg/(kg·d)were administrated by gavage according to specific groups.The behavioral tests were conducted after 4 weeks.At the same time,the expression of CAT and oxidative stress signal pathway was evaluated by Western Blot and immunohistochemistry.Results The AGO treatment for 4 weeks could significantly improve the depression like behavior of mice,at the same time,activate CAT in the hippocampus and serum of CRS mice,and alleviate oxidative stress injury.The DFX could partially offset this therapeutic effect.Conclusion The AGO can affect oxidative stress by changing CAT activity and thus play an antidepressant role.
