摘要
目的研究复方蜥蜴散凝胶通过调节胃癌前病变(PLGC)模型大鼠肠道内益生菌与致病菌平衡,修复损伤胃黏膜,抑制PLGC进展。方法将120只SD雄性大鼠随机分成空白对照组20只,模型组100只,采用甲基硝基亚硝基胍(MNNG)复合造模法制备模型。大鼠胃组织取材后,采用苏木素-伊红(HE)病理检测分析黏膜组织情况,对收集的粪便样本进行16S rRNA基因测序分析,采用R语言对菌群与通路进行关联性分析。结果复方蜥蜴散凝胶治疗后发现7个菌属发生了明显变化,分别是阿克曼菌属、类杆菌属、粪球菌属、乳杆菌属、普雷沃菌属、瘤胃球菌属、苏黎世杆菌属。结论复方蜥蜴散凝胶可通过对肠道菌群的双向调节作用如增殖有益菌,抑制有害菌,平衡肠道菌群结构,治疗PLGC。
Objective To study the compound lizard powder gel by adjusting the balance of probiotics and pathogenic bacteria in the intestines of rats model with gastric precancerous lesions(PLGC),repairing the damaged gastric mucosa,and inhibiting the progression of PLGC.Methods 120 SD male rats were randomly divided into blank control group(20 rats)and model control group(100 rats).The model was prepared by N-methyl-N′-nitro-N-nitroso-guani dine(MNNG)compound modeling method.After the rat gastric tissue was taken,the mucosal tissue was analyzed by hematoxylin-eosin(HE)pathological examination,the collected stool samples were sequenced and analyzed by 16 S rRNA gene,and the correlation between the flora and the pathway was analyzed by the R language.Results After the compound lizard powder gel treatment,obvious changes were found in 7 bacterial genera,namely akkermansia,bacteroides,coprococcus,lactobacillus,prevotella,ruminococcus and turicibacter.Conclusions The compound lizard powder gel could treat PLGC through the two-way regulation of gut microbiota,such as proliferation of beneficial bacteria,inhibiting harmful bacteria,balancing the structure of gut microbiota.
