摘要 介绍了两种常见的用于荧光定量PCR仪的温度控制系统。首先概述了采用经典的比例-积分-微分控制算法的温度控制系统,结合了自动调谐算法,具有易实现和循环时间短等优点,但是难以适应荧光定量PCR仪器高准确度的要求。因此,又介绍了一种荧光定量PCR仪器的多模态温度控制系统,采用了模糊PID控制算法,与第一种温度控制系统相比,该系统具有较短的稳态时间、较小的超调量、较强的鲁棒性和较高的准确度。介绍的两种温度控制系统对进一步开发利用荧光定量PCR仪的测试校准工作具有一定的参考价值。 In order to better carry out the metrological testing of fluorescent quantitative PCR instruments,it is necessary to understand the working principle of fluorescent quantitative PCR instruments,especially the principle of temperature control.This paper describes the common temperature control systems used for fluorescence quantitative PCR instruments.First,a temperature control system using the classical Proportional-Integral-Derivative(PID) control algorithm is outlined,which combines an auto-tuning strategy with the advantages of easy implementation and short cycle times,but is difficult to adapt to the high precision requirements of fluorescence PCR instruments.Therefore,a multimodal temperature control system for fluorescence quantitative PCR instruments is presented with a fuzzy PID control algorithm,which has shorter steady-state time,smaller overshoot,stronger robustness and higher accuracy than the previous temperature control system.The two temperature control systems introduced are of reference value for further development of testing and calibration of fluorescence quantitative PCR instruments.
