抗体芯片应用于真菌毒素多重检测的综合性实验设计

摘要 为践行科教协同育人理念,在病原生物学与免疫学实验教学环节,该文设计了抗体芯片应用于真菌毒素多重检测的综合性实验。基于竞争反应模式,通过设计抗体芯片布局和配套装置,同时制备真菌毒素完全抗原辣根过氧化物酶标物,经优化相关反应条件,最终建立了可实现真菌毒素多重检测的抗体芯片技术。结果显示,该方法可实现对4种真菌毒素的同时定量检测,灵敏度高、稳定性好。此综合性实验将病原微生物中真菌毒素的防控需求与免疫检测技术研究热点相结合,可在加深学生对课堂知识理解和掌握的同时,提升学习的全面性和系统性。与传统验证性实验相比,融入科研成果的综合性实验的开展,将巩固学生理论知识和实验操作技能,培养和激发其创新思维和科研热情,助力综合素质提升。 In order to practice the concept of science education cooperation,in the experimental teaching of pathogenic biology and immunology,a comprehensive experimental project is designed for the optimization and application of antibody microarray in the multiple mycotoxins determination.Based on the principle of competitive reaction mode,the antibody microarray layout and reaction hardware devices are designed,as well as the horseradish peroxidase labeled mycotoxin complete antigen is prepared.After optimizing the relevant reaction conditions,the antibody microarray that can realize multiple detection of mycotoxins is established.The results show that this immunological detection method can realize the simultaneous quantitative detection of four mycotoxins with high sensitivity and stability.This comprehensive experiment combines the prevention and control needs of mycotoxins with research hotspots of immune detection technology,which can deepen students’understanding and mastery of classroom knowledge and enhance the comprehensiveness and systematisms of learning.Compared with confirmatory experiments,this comprehensive experiments integrated with scientific research achievements can consolidate students’theoretical knowledge and experimental operation skills,cultivate and stimulate their innovative thinking and research enthusiasm,improve their comprehensive quality.
