电针调控大脑初级运动皮层第五层谷氨酸能神经元活性改善急性心肌缺血小鼠心功能的机制 被引量:2

Mechanism of electroacupuncture regulating the activity of glutamatergic neurons in the fifth layer of primary motor cortex to improve cardiac function in mice with acute myocardial ischemia

摘要 目的:观察电针对大脑初级运动皮层第五层谷氨酸能神经元(M1L5~(Glu))活动的影响,探究电针通过初级运动皮层改善心脏功能的神经调控机制。方法:将雄性C57BL/6小鼠随机分为伪手术组、模型组、电针组,每组7只,采用冠状动脉左旋前降支结扎的方法复制心肌缺血(MI)模型,电针组于造模后连续6 d给予电针双侧“神门”“通里”,20 min/次,每日1次。通过powerlab标准Ⅱ导联采集心电图信号,LabChart 8计算心率、ST段偏移,PhysioZoo分析心率变异性(HRV);HE染色法观察心肌细胞病理变化;病毒示踪法探究大脑皮层区心脏上游神经元集群分布,光纤光度法记录M1L5~(Glu)神经元活性变化;光遗传手段操控M1L5~(Glu)神经元观察心脏自主神经活动变化(LF/HF)。结果:与伪手术组比较,模型组小鼠5 min心电图LF/HF比值增高(P<0.01)且室性早搏(PVCs)发生增多;与模型组比较,电针组5 min心电图LF/HF比值降低(P<0.05)且PVCs发生减少,与伪手术组比较,模型组心肌组织的损伤程度明显加重;与模型组比较,电针组心肌组织的损伤程度有所改善。心脏上游大脑初级运动皮质区EGFP阳性神经元集群主要分布于M1L5区,且左右半球分布一致;与伪手术组比较,模型组100 s时间段内ΔF/F均值增加(P<0.01),钙事件增多(P<0.01);与模型组比较,电针组100 s时间段内ΔF/F均值降低(P<0.05),钙事件减少(P<0.05)。光遗传激活M1L5~(Glu)神经元,与空载病毒对照组比较LF/HF增加(P<0.01)。结论:电针可改善MI小鼠心肌损伤,减少PVCs的发生,其机制可能与降低M1L5~(Glu)神经元活性,进而调节心脏自主神经平衡有关。 Objective:To observe the effects of electroacupuncture on the activity of glutamatergic neurons(M1L5Glu)in the fifth layer of primary motor cortex and to explore the neural regulation mechanism of electroacupuncture in improving cardiac function through primary motor cortex.Methods:Male C57BL/6 mice were randomly divided into sham operation group,acute myocardial ischemia model group and electroacupuncture heart meridian group,with 7 mice in each group.The MI model was established by ligation of the left anterior descending branch of the coronary artery.The electroacupuncture heart meridian group was treated with electroacupuncture on both sides of‘Shenmen’and‘Tongli’for 6 days,20 min a day.ECG signals were collected by powerlab standardⅡleads,heartrate and HT height were calculated by LabChart8,heart rate variability(HRV)was analyzed by,PhysioZoo,pathological changes of cardiomyocytes were observed by HE staining,the distribution of upstream cardiac neurons in cerebral cortex was studied by virus tracing,and the activity changes of M1L5Glu neurons were recorded by optical fiber photometry.M1L5Glu neurons were manipulated by photogenetic means to observe the changes of cardiac autonomic nervous activity(LF/HF).Results:Compared with the sham operation group,the LF/HF ratio of 5 min electrocardiogram and the occurrence of ventricular extrasystole increased in the model group(P<0.01);compared with the model group,the LF/HF ratio of 5 min electrocardiogram(P<0.05)and the occurrence of ventricular extrasystole(PVCs)in the electroacupuncture heart meridian group decreased;compared with the sham operation group,the degree of myocardial injury in the model group was significantly aggravated;compared with the model group,the myocardial tissue injury in the electroacupuncture heart meridian group was improved.The clusters of EGFP positive neurons in the primary motor cortex of the upstream heart were mainly distributed in the M1L5 area,and the distribution was consistent between the left and right hemispheres.Com
