附子粗多糖的结构特征及抗氧化活性研究 被引量:2

摘要 目的:探究附子粗多糖的结构特征及抗氧化作用。方法:采用水提醇沉、三氯甲烷脱蛋白等方法制备附子粗多糖,通过紫外-可见光谱、热重分析以及扫描电镜推测其可能存在的结构,最后测定其DPPH自由基清除能力和羟自由基清除能力以评估多糖的抗氧化效果。结果:紫外光谱分析附子粗多糖不含核酸、蛋白质以及多肽等物质;扫描电镜观察其呈片状不规则形态;热重数据分析附子多糖具有两个失重阶段,具有典型的多糖类失重现象;在体外抗氧化活性研究中,附子粗多糖对DPPH和羟自由基的清除率分别高达98.01%和96.08%,具有较强的体外抗氧化活性。结论:附子粗多糖热稳定较好,抗氧化能力较强,可作为潜在的抗氧化剂。 Objective:To explore the structural characteristics and antioxidant effects of Fuzi polysaccharides.Method:The polysaccharides were prepared by water extraction,alcohol precipitation and trichloromethane deproteinization methods.The possible structure of the polysaccharides was inferred by UV-vis spectrum,thermogravimetric analysis and scanning electron microscope.Finally,the DPPH radical and hydroxyl radical scavenging abilities were determined to evaluate the antioxidant effects of the polysaccharides.Result:UV spectrum analysis showed that Fuzi polysaccharides contained no nucleic acid,protein and polypeptides.Scanning electron microscope displayed its irregular flake shape.Thermogravimetric data analysis showed that Fuzi polysaccharides had two weightlessness stages,with typical polysaccharide weightlessness phenomena.In the in vitro antioxidant activity study,the scavenging rates of DPPH and hydroxyl radical by Fuzi polysaccharides were up to 98.01%and 96.08%,respectively,having strong in vitro antioxidant activity.Conclusion:Fuzi polysaccharides have good thermal stability and strong antioxidant abilities,which can be used as a potential antioxidant.
