基于TRPC6通道探讨针刺介导缺血性脑卒中神经、血管保护的研究进展

摘要 TRPC6通道与缺血性脑卒中密切相关,在缺血、缺氧等应激条件下对维持细胞存活和神经保护起着关键作用。TRPC6通道作为非选择性阳离子通道,调控卒中后细胞内外Ca^(2+)的变化,参与缺血后一系列病理生理过程。研究发现,针刺可以调控TRPC6通道相关因子的表达如:血管内皮生长因子VEGF、脑源性神经生长因子BDNF、钙调素依赖性蛋白激酶ⅡCAMKⅡ、钙调素依赖性蛋白激酶ⅣCAMKⅣ、蛋白激酶A PKA、白细胞介素IL与抗凋亡因子Bcl-2。与此同时,TRPC6通道作为“针刺反应通道”也可以感知CREB途径整合的针刺信息以改善缺血性脑卒中的预后。本研究基于TRPC6通道对缺血性脑卒中内源性保护机制及针刺干预作用进行探析,以期为针刺治疗缺血性脑血管病提供新参考。 Canonical transient receptor potential 6 channel(TRPC6)is closely associated with ischemic stroke and plays a key role in maintaining cell survival and neuroprotection under stress conditions of ischemia and hypoxia.As a non-selective cation channel,the TRPC6 channel regulates the change of intracellular and extracellular Ca 2+after stroke and participates in a series of pathophysiological processes of post-ischemia.It has been found that acupuncture can regulate the expression of TRPC6 channel-related factors,including vascular endothelial growth factor(VEGF),brain-derived neurotrophic factor(BDNF),and calcineurin-dependent protein kinase II(CAMKII),calmodulin-dependent protein kinase IV(CAMKIV),protein kinase A(PKA),interleukin IL,and anti-apoptotic factor Bcl-2.At the same time,the TRPC6 channel,as a‘acupuncture response channel’,can also perceive the acupuncture information integrated by the CREB pathway to improve the prognosis of ischemic stroke.Based on the TRPC6 channel,the endogenous protection mechanism of ischemic stroke and the intervention effect of acupuncture are explored to provide a new reference for acupuncture in treatment of ischemic cerebrovascular diseases.
