摘要
伴随广谱抗生素的过度使用,临床上多重耐药病原菌和超级细菌不断出现,为临床抗感染治疗带来了严峻挑战,传统微生物培养后药敏实验以及针对于特定耐药基因的基因扩增等方法已经无法满足现有需要,因而迫切需要新的研究方法的出现。宏基因组测序技术以其无偏差、高覆盖为微生物耐药研究提供了又一有利工具,本文对宏基因组测序在肠道、皮肤、组织和重症下呼吸道感染中微生物耐药研究的最新应用做一综述,以期为临床微生物耐药研究提供帮助。
With the overuse of broad-spectrum antibiotics,multidrug-resistant pathogens and superbugs emerge in clinic,severe challenges have been created in clinical treatment.Traditional microbial culturing antibiotic susceptibility tests and gene amplification test for specific antibiotic resistant genes were not able to meet the existing needs,suggesting the new research methods are urgently needed.Metagenomic sequencing technology provides an alternative valuable tool for microbial resistant gene research with its unbiased feature and high coverage.This paper reviews the latest applications of metagenomic sequencing in microbial antibiotic resistant research in gut,skin,tissue and severe lower respiratory tract infections,in order to provide useful knowledge for clinical microbial antibiotic resistance research.
