子宫内膜癌中血清反应因子家族表达特征和免疫细胞浸润的综合分析 被引量:1

Comprehensive analysis of serum response factor family expression characteristics and immune cell infiltration in the endometrial carcinoma

摘要 目的探讨转录因子血清反应因子(SRFs)家族成员在子宫内膜癌中的差异表达、生物学功能、肿瘤免疫浸润和临床预后价值。方法以癌症基因组图谱(TCGA)数据库为基础,用R 3.6.3进行差异表达基因分析、基因集富集分析和预后分析;用Metascape进行基因本体(GO)分析;用pROC包构建受试者工作特征(ROC)曲线;用cBioPortal数据库进行基因突变、基因相关性分析;用人类基因图谱数据库进行蛋白质表达分析;用基因集变异分析包进行免疫浸润分析。结果子宫内膜癌中肌细胞增强因子2BNB-肌细胞增强因子2B(BORCS8-MEF2B)、肌细胞增强因子2A(MEF2A)、肌细胞增强因子2C(MEF2C)、肌细胞增强因子2D(MEF2D)和SRF的表达均显著低于正常子宫内膜组织(P<0.001);SRF在Ⅰ期和Ⅲ期之间表达差异有统计学意义(P<0.05),而SRFs家族其余成员的表达在子宫内膜癌各临床分期之间差异无统计学意义(P>0.05);SRFs家族成员的表达与子宫内膜癌的总生存率、疾病特异性生存率和疾病无进展间期均无相关性(P>0.05);ROC曲线分析显示,BORCS8-MEF2B、MEF2A、MEF2C、MEF2D和SRF均具有诊断价值,曲线下面积分别为0.741、0.936、0.987、0.907和0.931;MEF2A的表达与MEF2C的表达呈正相关,MEF2D的表达与SRF的表达呈正相关,其他各SRFs家族成员之间无相关性;111个基因与SRFs家族6个基因显著密切相关;GO分析显示,这些基因与hsa03040、GO:0005681、GO:0030687、GO:0071013和GO:0008023等条目相关;这些基因相关的基因通路包括hsa03040、hsa04010、hsa04022、hsa04371、hsa04928、hsa05418等;免疫浸润分析显示SRFs家族成员与免疫细胞浸润的关系密切。结论BORCS8-MEF2B、MEF2A、MEF2C、MEF2D和SRF表达水平的改变可能在子宫内膜癌的发生、发展中起重要作用,可能成为子宫内膜癌的诊断指标。 Objective To investigate the differential expression,biological function,tumor immune invasion and clinical prognostic value of serum response factors(SRFs)family members in endometrial cancer.Methods Based on The Cancer Genome Atlas(TCGA)database,R 3.6.3 was used to analyze differentially expressed genes,gene set enrichment analysis and prognosis analysis.Gene ontology(GO)analysis was done by using Metascape.The receiver operating characteristic(ROC)curve was constructed by using pROC package.Gene mutation and gene correlation were analyzed by cBioPortal database.The Human Protein Atlas database was used for protein expression analysis.Immune infiltration was analyzed by gene set variation analysis package.Results The expressions of myocyte enhancer factor 2BNB-myocyte enhancer factor 2B(BORCS8-MEF2B),myocyte enhancer factor 2A(MEF2A),myocyte enhancer factor 2C(MEF2C),myocyte enhancer factor 2D(MEF2D)and SRF in endometrial cancer were significantly lower than those in normal endometrial tissues(P<0.001).There was no significant difference in the expression of SRFs family members in different clinical stages of endometrial cancer(P>0.05).Except SRF expression between stageⅠand stageⅢ(P<0.05),there was no correlation between the expression of SRFs family members and overall survival,disease specific survival and progression-free interval of endometrial cancer(P>0.05).ROC analysis showed that BORCS8-MEF2B,MEF2A,MEF2C,MEF2D and SRF had diagnostic value,and the areas under the curve were 0.741,0.936,0.987,0.907 and 0.931,respectively.The expression of MEF2A was positively correlated with the expression of MEF2C,and the expression of MEF2D was positively correlated with the expression of SRF.There was no significant correlation between other SRFs family members.111 genes were significantly correlated with 6 genes of SRFs family.GO analysis showed that these genes were related to hsa03040,GO:0005681,GO:0030687,GO:0071013 and GO:0008023.The gene pathways related to these genes included hsa03040,hsa04010,hsa04022,hsa
