摘要
本研究以粉皮高油酸花生品系‘G94’与紫皮常规花生品系‘黑珍珠’为亲本配置杂交组合以期创制具有较高医疗保健功能的紫色种皮高油酸花生种质新材料。利用等位基因特异性PCR(allele-specific PCR,AS-PCR)方法,采用ahFAD2A/ahFAD2B引物筛选F_(2)群体中_ol_(1)_ol_(2)单株。F_(2:3)家系种皮色泽χ^(2)检测结果表明,该组合双亲在种皮色泽性状上存在1对差异基因,种皮紫色对粉色为完全显性。利用AS-PCR方法鉴定ol_(1)ol_(1)ol_(2)ol_(2)基因型的紫色种皮F_(3)单株,通过系谱选育,获得遗传稳定的紫色种皮高油酸新种质4份,9-6-2-7、9-6-2-8、9-6-16-5和9-6-18-1。单株荚果重较对照‘冀花2号’分别提高180.3%、171.9%、203.6%和178.3%,株高分别降低13.35%、23.29%、42.93%和25.16%。经气相色谱测定,该批新种质的油酸GC值介于80.77%~82.87%,O/L介于18.91~29.49。这些新种质不仅油酸含量高于75%,而且种皮内富含花青素类物质,对丰富中国高油酸花生种质资源遗传多样性具有重要的现实意义。
High oleic acid line'G94'with pink testa and normal oleic acid line'Heizhenzhu'with purple testa were used as parents in order to create new germplasm materials combined with purple testa and high oleic acid peanut with higher medical and health functions.The genetype_ol_(1)_ol_(2)individuals were screened via allele-specific PCR(AS-PCR)method with ahFAD2A/ahFAD2B primer in F_(2)plants.Genetic rule was inferred based on F_(2:3)family lines phenotype proportion through chi-squest test.The results showed that peanut purple testa color trait was controlled by one pairs of different expressed genes between the two parents,which revealed seed testa purple was completely dominant gene.Purple testa peanut with ol_(1)ol_(1)ol_(2)ol_(2)genetype were identified and selected in F_(3)plant using the AS-PCR method.After successive transfer breeding,4 genetic stable germplasm materials with purple testa and oleic acid,which is including 9-6-2-7,9-6-2-8,9-6-16-5 and 9-6-18-1,were obtained.Compared to control'Jihua 2',pod weights per plant were increased 180.3%,171.9%,203.6%and 178.3%respectively and the height of plant were decreased 13.35%,23.29%,42.93%and 25.16%respectively.The result of determination of gas chromatography indicated that the oleic acid GC value was between 80.77%~82.87%and O/L was 18.91~29.49.These new germplasms not only contain more than 75%oleic acid,but are also rich in anthocyanins in testa,which has important practical significance for enriching the genetic diversity of high oleic peanut germplasm resources in China.
