摘要
松材线虫病是一种林业危害性流行病,传播迅速、危害巨大,对中国在内的多个国家造成了严重的生态和经济损失。松材线虫和拟松材线虫作为中国松材线虫病的病原线虫,依靠媒介昆虫在松科植物传播,目前其防治仍存在困难。鉴别该病的病原线虫松材线虫与拟松材线虫一直是研究的热点,目前主流的鉴定方法分为形态检测、生化检测与分子检测3大方向。绝大多数传统的松材线虫检测技术存在一些局限性,如特异性较差、灵敏度不高、基层推广难度较大等;而基于核酸的分子检测技术以其简单,快捷,灵敏,可靠的优点逐渐成为检测研究的主流方向。从形态、生化分子、核酸检测3方面综述了3类松材线虫鉴定方法,并比较了各鉴定方法的优缺点,旨在为探索新的松材线虫病的鉴定方法提供理论依据,为研究松材线虫病提供新的方向。
Pine wood nematode disease is a kind of harmful epidemic in forestry,which spreads rapidly and does great harm,and has caused serious ecological and economic losses to many countries,including China.As the pathogenic nematodes of pine wood nematode disease in China,Bursaphelenchus xylophilus and B.mucronatus rely on vector insects to spread in pines,but there are still difficulties in control them.The identification of the pathogenic nematodes of B.xylophilus and B.mucronatus has always been a focus of research.Currently,the identification methods of B.xylophilus and B.mucronatus are mainly divided into three directions:morphological identification,biochemical identification and molecular identification.Most traditional identification techniques in pine wood nematodes have some limitations,such as poor specificity,low sensitivity,and difficult promotion at the grass-roots level.The molecular identification technology based on nucleic acid has gradually become the main direction of detection research because of its advantages of simple,fast,sensitive and reliable features.In this paper,three identification methods were reviewed from the aspects of morphology,biochemical molecules and nucleic acid identification.The advantages and disadvantages of each identification method were compared,in order to provide a theoretical basis for the exploration of new identification methods for pine wood nematode diseases,and provide a new direction for the study of pine wood nematode diseases.
