摘要
[目的]本研究旨在探讨大蒜辣素对四氯化碳(CCl_(4))诱导的小鼠急性肝损伤的缓解作用及其潜在作用机制。[方法]将42只小鼠随机分为7组,分别为对照组、大蒜辣素处理组、CCl_(4)处理组、复方甘草酸苷治疗组(80 mg·kg^(-1))及3个大蒜辣素(40、20和10 mg·kg^(-1))治疗组。小鼠腹腔注射1%CCl_(4)(10 mL·kg^(-1))8 h后灌胃给予不同剂量的大蒜辣素和复方甘草酸苷,于CCl_(4)处理48 h后采集小鼠血液和肝脏组织,测定血清生化指标、血清中炎性因子以及肝组织MDA及CAT含量,同时采用Western blot方法检测大蒜辣素对细胞凋亡相关蛋白表达的影响。[结果]单独使用大蒜辣素对小鼠肝脏无损伤作用(P>0.05)。40、20和10 mg·kg^(-1)大蒜辣素均可显著降低CCl_(4)处理小鼠的血清AST、ALT及ALP活性水平(P<0.05),显著提高肝脏CAT含量和降低MDA含量(P<0.05)。40 mg·kg^(-1)大蒜辣素还可显著降低CCl_(4)处理小鼠血清炎性因子TNF-α、IL-1β和IL-6水平(P<0.05)以及肝组织Bax、Cleaved-Caspase 3和Cleaved-Caspase 9蛋白表达水平(P<0.05),显著提高肝组织NQO_(1)和Nrf_(2)蛋白表达水平(P<0.05)。在降低小鼠血清生化指标、炎性因子和MDA水平方面,高、中剂量的大蒜辣素优于复方甘草酸苷(P<0.05)。[结论]大蒜辣素可通过抗氧化、抗炎作用以及减少肝细胞凋亡对CCl 4引起的急性肝损伤起到缓解作用,有望成为新型保肝药物在兽医临床中应用。
[Objectives]The aim of this study was to investigate the alleviating effect of allicin on acute liver injury caused by carbon tetrachloride(CCl_(4)) in mice and its potential mechanism.[Methods]Fourty-two mice were randomly divided into 7 groups:control group,allicin treatment group,CCl_(4) treatment group,compound glycyrrhizin treatment group(80 mg·kg^(-1)),and different doses of allicin group(40,20 and 10 mg·kg^(-1)).Mice were intraperitoneally injected with 1%CCl_(4)(10 mL·kg^(-1)body weight)for 8 h,then given different doses of allicin and compound glycyrrhizin by intragastric administration.After 48 h of CCl_(4) treatment,blood and liver tissues of mice were harvested,and serum biochemical indices,inflammatory factors in serum and the contents of MDA and CAT in liver tissue were measured.Western blot was used to detect the effect of allicin on apoptosis related protein expression.[Results]Allicin alone had no damage to liver of mice during the experiment(P>0.05).Allicin at 40,20 and 10 mg·kg^(-1) body weight significantly decreased the serum of AST,ALT and ALP levels in CCl_(4)-treated mice(P<0.05),significantly increased the content of CAT and inhibited the content of MDA in liver(P<0.05).In addition,40 mg·kg^(-1)body weight allicin significantly reduced the serum TNF-α,IL-1βand IL-6 levels of CCl_(4)-treated mice(P<0.05),and liver protein levels of BAX,Cleaved-Caspase 3 and Cleaved-Caspase 9,NQO_(1) and Nrf_(2)(P<0.05).The effects of high and medium doses of allicin on serum biochemical indexes,inflammatory factors and MDA content in mice were better than those of compound glycyrrhizin(P<0.05).[Conclusions]The results indicated that allicin alleviated CCl_(4)-induced acute liver injury via the modulation of oxidative stress,inflammation,and apoptosis associated with progress of liver damage,which could be a promising hepatoprotective agent used in veterinary clinic.
