基于“表里关联”和化学计量学的西洋参微波炮制过程中皂苷含量与色度变化的分析 被引量:2

Analysis on the Changes in Contents of Saponins and Chromatic Values in American Gingseng During Microwave Processing Based on “Surface in Relation” and Stoichiometry

摘要 目的通过测定西洋参在微波炮制过程中人参皂苷含量与色度值的变化并研究其关联性,为西洋参微波炮制品种的探索和质量标准的制定提供数据支持。方法采用高效液相色谱法测定不同炮制程度的西洋参样品中11种人参皂苷的含量,采用精密色差仪测定不同炮制程度饮片粉末的色度值;运用中药色谱指纹图谱相似度评价系统(2012年版)建立不同炮制程度饮片的HPLC指纹图谱;SPSS 21.0软件对样品的色度值与皂苷含量进行关联性分析并进行聚类分析,采用化学计量学方法对结果进行分析。结果HPLC结果显示随炮制程度加深皂苷种类及含量均有变化;指纹图谱共标定28个共有峰,指认了其中7个色谱峰,发现2个峰为炮制后新产生,2个峰在炮制后消失;相关性结果显示,大部分共有峰及皂苷类成分与L*值、a*值及E*值呈显著相关性;化学计量学结果显示,17批样品可分为4类,其中人参皂苷Rb1和Re为炮制过程中变化的差异性成分。结论西洋参在微波炮制过程中色度值与皂苷含量均发生显著变化,且两者有一定的表里关联性,西洋参微波炮制具有一定可行性,值得进一步研究与探索。 Objective The changes of ginsenoside contents and chromatic values of Panax quinquefolium during microwave processing were determined and their correlation was studied,which provided data support for the exploration of microwave processing varieties and the establishment of quality standard of Panax quinquefolium.Methods The contents of 11 ginsenosides in panax quinquefolium samples with different processing degrees were determined by HIGH performance liquid chromatography,and the chromaticity values of powder with different processing degrees were determined by precision chromaticity meter.“Chinese Medicine Chromatographic Fingerprint Similarity Evaluation Software(2012 Version)” was used to establish HPLC fingerprint of decoction pieces with different processing degrees.SPSS 21.0 software was used for correlation analysis and cluster analysis of chromatic values and ginsengoside contents of samples,the results were analyzed by stoichiometry.Results HPLC results showed that the varieties and contents of ginsengosides changed with the deepening of processing degree.A total of 28 common peaks were identified by fingerprint,and 7 chromatographic peaks were identified.Two peaks were newly generated after processing,and two peaks disappeared after processing.Correlation results showed that most of the common peaks and saponins were significantly correlated with L*,A * and E* values.The stoichiometry results showed that the 17 batches of samples could be divided into four categories,among which ginsenoside Rb1 and Re were the different components in the process of processing.Conclusion The chromatic values and saponin contents of Panax quinquefolium were significantly changed during microwave processing,and there was a certain correlation between the two,which indicated that microwave processing of Panax quinquefolium was feasible and worthy of further study and exploration.
