黄芪提取物对病毒性心肌炎感染模型大鼠氧化应激及心功能保护的影响 被引量:2

Effect of astragalus extract on oxidative stress and cardiac function protection on rat models with viral myocarditis infection

摘要 目的分析黄芪提取物对病毒性心肌炎感染大鼠的疗效,并探究其对于大鼠氧化应激及心功能保护的作用机制。方法选取SPF级Wistar大鼠50只,10作为对照组,其余40只建立心肌炎感染模型,选取建模成功的30只大鼠随机平均分为模型组、黄芪提取物低、高剂量组。观察大鼠病理评分、心功能指标[左心室收缩末期内径(LVESD)、左心室舒张末期内径(LVEDD)、左室射血分数(LVEF)]、炎性因子[白细胞介素(IL)-6、IL-10和肿瘤坏死因子-α(TNF-α)]、氧化应激[超氧化物歧化酶(SOD)和丙二醛(MDA)]和通路蛋白[核因子红细胞系2相关因子2(Nrf2)、血红素加氧酶1(HO-1)]表达。结果与对照组比较,模型组、黄芪提取物低、高剂量组LVESD、LVEDD、IL-6、TNF-α、IL-10、MDA表达水平升高,LVEF、SOD、Nrf2、HO-1表达水平降低(P<0.05);与模型组比较,黄芪提取物低、高剂量组病理评分、LVESD、LVEDD、IL-6、TNF-α、IL-10、MDA表达水平降低,LVEF、SOD、Nrf2、HO-1表达水平升高(P<0.05);与黄芪提取物低剂量组相比,黄芪提取物高剂量组病理评分、LVESD、LVEDD、IL-6、TNF-α、IL-10、MDA表达水平降低,LVEF、SOD、Nrf2、HO-1表达水平升高(P<0.05)。结论黄芪提取物可以抑制病毒性心肌炎感染大鼠心肌细胞损伤,改善氧化应激水平,其作用机制可能与Nrf2、HO-1通路有关。 OBJECTIVE To observe the effect of astragalus extract on oxidative stress and cardiac function protection of the rat models with viral myocarditis infection.METHODS Totally 50 SPF Wistar rats were chosen as research subjects,10 of which were assigned as the control group,and the rest of 40 rats were selected to establish the myocarditis infection models.30 rat models were randomly divided into the model group,the low-dose astragalus extract group and the high-dose astragalus extract group.The pathological score,cardiac function indexes[left ventricular end-systole diameter(LVESD),left ventricular end diastolic dimension(LVEDD),left ventricular ejection fraction(LVEF)],inflammatory factors[interleukin(IL)-6,IL-10,tumor necrosis factor-α(TNF-α)],oxidative stress[superoxide dismutase(SOD),malondialdehyde(MDA)]and pathway proteins[nuclear factor erythron 2-related factor(Nrf2),heme oxygenase 1(HO-1)]were observed and compared among the groups.RESULTS The LVESD,LVEDD,IL-6,TNF-α,IL-10 and MDA expression levels of the model group,the low-dose astragalus extract group and the high-dose astragalus extract group were higher than those of the control group,while the LVEF,SOD,Nrf2 and HO-1 expression levels of the model group,the low-dose astragalus extract group and the high-dose astragalus extract group were lower than those of the control group(P<0.05).The pathological score,LVESD,LVEDD,IL-6,TNF-α,IL-10 and MDA expression levels of the low-dose astragalus extract group and the high-dose astragalus extract group were lower than those of the model group,while the LVEF,SOD,Nrf2 and HO-1 expression levels of the low-dose astragalus extract group and the high-dose astragalus extract group were higher than those of the model group(P<0.05).The pathological score,LVESD,LVEDD,IL-6,TNF-α,IL-10 and MDA expression levels of the high-dose astragalus extract group were lower than those of the low-dose astragalus extract group,while the LVEF,SOD,Nrf2 and HO-1 expression levels of the high-dose astragalus extract group were higher t
