非小细胞肺癌患者血清长链非编码RNA LOC285194和微小RNA⁃211的表达水平及其临床意义 被引量:4

Expression levels of serum long non⁃coding RNA LOC285194 and microRNA⁃211 in patients with non⁃small cell lung cancer and its clinical significance

摘要 目的探讨非小细胞肺癌(NSCLC)患者血浆长链非编码RNA(lncRNA)LOC285194和微小RNA(miR)⁃211的表达水平及其临床意义。方法纳入NSCLC患者46例作为NSCLC组,同期42例健康体检者作为对照组。采用实时荧光定量聚合酶链反应(qRT⁃PCR)检测两组受试者血清lncRNA LOC285194和miR⁃211表达水平并进行比较,同时比较不同临床特征NSCLC患者血清lncRNA LOC285194和miR⁃211表达情况。NSCLC患者血清lncRNA LOC285194与miR⁃211表达水平的相关性分析采用Pearson相关分析。采用受试者工作特征(ROC)曲线分析血清lncRNA LOC285194、miR⁃211表达水平单独及二者联合对NSCLC的诊断效能。结果NSCLC组受试者血清lncRNA LOC285194表达水平低于对照组,miR⁃211表达水平高于对照组(P<0.001)。有无肿瘤远处转移及淋巴结转移、不同肿瘤分期NSCLC患者血清lncRNA LOC285194、miR⁃211表达情况构成比比较差异均有统计学意义(P<0.05)。Pearson相关分析结果显示,NSCLC患者血清lncRNA LOC285194与miR⁃211表达水平呈负相关(r=-0.795,P<0.001)。ROC曲线分析结果显示,血清lncRNA LOC285194表达水平诊断NSCLC的ROC曲线下面积(AUC)为0.809(95%CI 0.709~0.909),敏感度为80.4%、特异度为74.7%;血清miR⁃211表达水平诊断NSCLC的AUC为0.805(95%CI 0.701~0.910),敏感度为84.8%、特异度为76.5%;二者联合诊断NSCLC的AUC为0.881(95%CI 0.805~0.957),敏感度为89.1%、特异度为77.2%。结论NSCLC患者血清lncRNA LOC285194表达水平降低,miR⁃211表达水平升高,二者呈负相关并与肿瘤远处转移、淋巴结转移、肿瘤分期密切相关,且均对NSCLC具有一定诊断价值,二者联合时诊断价值更高。 Objective To investigate the expression levels of serum long non⁃coding RNA(lncRNA)LOC285194 and microRNA(miR)⁃211 in patients with non⁃small cell lung cancer(NSCLC)and its clinical significance.Methods A total of 46 patients with NSCLC were selected as NSCLC group,and 42 healthy subjects were selected as control group during the same period.Real⁃time fluorescence quantitative polymerase chain reaction(qRT⁃PCR)was used to detect expression level of serum lncRNA LOC285194 and miR⁃211 in subjects of two groups and to compare the results,meanwhile,expression condition of serum lncRNA LOC285194 and miR⁃211 in patients with NSCLC of different clinical characteristics were compared.Relationship between expression levels of serum lncRNA LOC285194 and miR⁃211 in patients with NSCLC was analyzed by Pearson correlation analysis.Diagnostic efficacy of expression level of serum lncRNA LOC285194 and miR⁃211 alone and combination in NSCLC were analyzed by receiver operating characteristic(ROC)curve.Results Expression level of serum lncRNA LOC285194 in NSCLC group was lower than that in control group,and expression level of serum miR⁃211 was higher than that in control group(P<0.001).There were statistically significant differences in ratio of expression condition of serum lncRNA LOC285194 and miR⁃211 in NSCLC patients with or without distant metastasis,lymph node metastasis and different tumor stages(P<0.05).Pearson correlation analysis showed that expression level of serum lncRNA LOC285194 in patients with NSCLC was negatively correlated with miR⁃211(r=-0.795,P<0.001).ROC curve analysis showed that area under ROC curve(AUC)of expression level of serum lncRNA LOC285194 in diagnosis of NSCLC was 0.809(95%CI 0.709⁃0.909),sensitivity was 80.4%and specificity was 74.7%.AUC of expression level of serum miR⁃211 in diagnosis of NSCLC was 0.805(95%CI 0.701⁃0.910),sensitivity was 84.8%and specificity was 76.5%.AUC of expression level of serum lncRNA LOC285194 combined with miR⁃211 in diagnosis of NSCLC w
