摘要
为了探明褪黑激素(melatonin, MLT)埋植剂埋植不同时间对水貂睾丸形态和显微结构的影响,试验于MLT埋植剂埋植满158 d和248 d时,从埋植MLT的试验组(试验158组、试验248组)和未埋植的对照组(对照158组、对照248组)中随机各选取水貂3只,剖杀后取睾丸并称重,测定睾丸长径、宽径和厚径并计算体积;将睾丸制成石蜡切片,显微镜下观察形态学与组织学特征,测量生精小管直径并计算上皮细胞层数。结果表明:试验158组水貂睾丸长径、宽径、厚径、体积、重量、生精小管直径和上皮细胞层数均极显著高于对照158组(P<0.01),试验248组较对照248组和试验158组均极显著减少(P<0.01);试验158组与对照248组相比,除生精小管直径显著缩小(P<0.05)和上皮细胞层数极显著减少(P<0.01)外,其他均差异不显著(P>0.05);试验158组生精小管内细胞种类较对照158组多,分布更紧密,成熟精子比例和生精细胞数量较对照248组少;试验248组与对照248组相比生精细胞数量与种类更少,上皮细胞主要由精子细胞构成。说明埋植MLT埋植剂158 d可促进水貂睾丸发育,使体积与重量达到发情期状态,但生精小管与上皮细胞尚未完全发育;而埋植248天时,水貂睾丸各项指标均低于对照组,处于退化状态。
In order to explore the effects of melatonin(MLT)implant on morphology and histology of mink testes at different implantation times,in the experiment on 158d and 248 d of MLT implantation,three mink were randomly selected from each of the test groups(test group 158 and test group 248)and the control groups(control group 158 and control group 248)which not implanted with MLT.Testes were taken after dissection,the weight,the length,width and thickness of the testes were determined and the volume was calculated.Paraffin sections were made of the testes and morphological and histological features were observed under the microscope,and the diameter of the germinal tubules was measured and the number of epithelial cell layers was calculated.The results showed that the testicular length,width,thickness,volume,weight,diameter of germinal tubules and number of epithelial cell layers were all significantly higher in the test group 158 than those in the control group 158(P<0.01),and the test group 248 had a highly significant decrease compared with the control group 248 and the test group 158(P<0.01).Compared with the control group 248,the differences were not significant(P>0.05)in the test group 158,except for a significant decrease in the diameter of the germinal tubules(P<0.05)and a highly significant decrease in the number of epithelial cell layers(P<0.01);the test group 158 had more types of cells in the germinal tubules than the control group 158,which were more closely distributed,and the proportion of mature spermatozoa and the number of germinal cells were less than those in the control group 248;the number and type of spermatogonia were less in the test group 248 compared with the control group 248,and the epithelial cells were mainly composed of spermatozoa.The results suggested that on 158 d of MLT implantation,the testes of mink were developed and the volume and weight reached the state of estrus,but the growth of germinal tubules and epithelial cells were not fully developed;while on 248 d of implantation,all
