摘要
目的 ARID1A是SWI/SNF染色质复合体的关键成员,该基因已成为人类各种癌症的肿瘤抑制基因。分析了ARID1A在甲状腺癌中的表达情况和临床意义。方法 利用免疫组化技术对甲状腺癌患者组织与癌旁组织进行检测,并结合聚合酶链反应(polymerase chain reaction, PCR)技术了解ARID1A基因在甲状腺癌中的mRNA表达情况。此外,本研究还通过蛋白质印迹技术、MTT增殖试验、集落形成试验以及细胞凋亡检测技术探究ARID1A在甲状腺癌发生、发展中对肿瘤细胞增殖和凋亡影响的作用机制。结果 与正常腺上皮比较,甲状腺癌组织中ARID1A表达降低,并且与淋巴结转移、TNM分期和分化差具有显著的相关性,并且ARID1A在甲状腺癌细胞系中的表达低于正常腺上皮TEC细胞系。本研究还进一步探讨了ARID1A在甲状腺癌细胞系生物学行为中的作用,FTC133和BCPAP细胞系中转染小干扰RNA(siRNA)的ARID1A缺失促进增殖、集落形成能力,并抑制细胞凋亡。结论 ARID1A可能是甲状腺癌中的重要肿瘤抑制因子,该基因的下调能够促进甲状腺癌细胞增殖,抑制细胞凋亡。
Objective ARID1A is a key member of SWI/SNF chromatin complex,which has become a tumor suppressor gene in various human cancers.This study analyzed the expression and clinical significance of ARID1A in thyroid carcinoma.Methods Immunohistochemical technique was used to detect the mRNA expression of ARID1A gene in thyroid carcinoma and paracancerous tissues,and combined with polymerase chain reaction(PCR)technology to understand the mRNA expression of ARID1A gene in thyroid carcinoma.In addition,this study also explored the mechanism of the effect of ARID1A on the cell proliferation and apoptosis of tumor in the occurrence and development of thyroid carcinoma by Western blot,MTT proliferation test,colony formation test and apoptosis detection techniques.Results Compared with normal glandular epithelium,the expression of ARID1A in thyroid cancer tissues was reduced,and was significantly correlated with lymph node metastasis,TNM stage and poor differentiation,and ARID1A expression in thyroid cancer cell lines was lower than in normal glandular epithelial TEC cell lines.The role of ARID1A in the biological behavior of thyroid cancer cell lines was further investigated,and ARID1A deletion by small interfering RNA(siRNA)in FTC133 and BCPAP cell lines promoted proliferation,colony-forming ability,and inhibited apoptosis.Conclusion ARID1A may be an important tumor suppressor in thyroid cancer,and down-regulation of this gene can promote the proliferation of thyroid cancer cells and inhibit cell apoptosis.
