产类胡萝卜素酵母的筛选及全基因组分析

摘要 锁掷酵母是一类可以产生多种类胡萝卜素、安全性较高的微生物,近年来得到广泛关注。试验对筛选得到的产类胡萝卜素酵母进行产量及生物学特性测定,通过全基因组分析对Sporidiobolus pararoseus TR71菌株安全性进行评估并探究其潜在的益生特性,并采用DMSO法测定类胡萝卜素产量。通过Illumina NovaSeq平台进行全基因组测序。结果表明:所筛选酵母类胡萝卜素平均产量为1.65 mg/L,最高产量为4.27 mg/L。菌株S. pararoseus TR71具有蛋白酶及脲酶活性,基因组序列全长为13 698 066 bp,CG含量为47.36%,有多个基因基因被KEGG、eggNOG等多个数据库注释。试验分离得到的S. pararoseus TR71菌株有相对较高的安全性,有希望成为饲喂益生菌制剂的后备菌株。 Sporidiobolus is a kind of microorganism that can produce many carotenoids with high safety,which has attracted a lot of attention in recent years.The yield and biological characteristics of the screened carotenoid-producing yeast were determined,the safety of the Sporidiobolus pararoseus TR71 strain was evaluated by genome-wide analysis,and its potential probiotic properties were explored.The carotenoid yield was determined by the DMSO method.The whole genome was sequenced by the Illumi⁃na NovaSeq platform.The results showed that the average carotenoid yield was 1.65 mg/L and the high⁃est yield was 4.27 mg/L.The strain S.paaroseus TR71 had protease and urease activities.The total length of the genome sequence was 13698066 bp,the CG content was 47.36%,and several genes were annotated by KEGG,eggNOG,and other databases.S.pararoseus TR71 strain isolated from the ex⁃periment has relatively high safety,and it is promising to be a reserve strain for feeding probiotics.
