基于粪便激素检测的圈养赤斑羚发情周期初探 被引量:1

Estimation of Reproductive Cycle of Naemorhedus baileyi Based on Fecal Hormone Analysis

摘要 采集上海动物园赤斑羚(Naemorhedus baileyi)的粪便,探讨不同粪样保存方法对生殖激素检测结果的影响,并应用粪便生殖激素水平预测赤斑羚的发情周期。结果表明:干燥和冷冻对于雌二醇的保存效果较好;乙醇浸泡和冷冻对孕酮的保存效果较好;冷冻对睾酮的保存效果较好;抗生素保存仅在15 d以内有效。综合考虑各方法,冷冻保存的效果最好,乙醇浸泡适合短期样本运输及保存,干燥法作为没有冷冻条件时的替代法。2018年1月—2019年12月的粪便生殖激素测定表明,雌二醇于2018年1月上旬及12月出现波峰,2019年2—4月水平较高,10—12月出现高峰;孕酮在2018年1月、11—12月中旬出现多个峰值,2019年1月出现波峰,2月保持高水平,10—12月中旬持续波动;睾酮水平在2018年11月—2019年2月,以及10月和11月出现峰值。综合考虑所测数据,初步预测雌性赤斑羚发情期为10月—翌年3月末,其中12月—翌年1月为发情高峰期,雄性赤斑羚发情期为10月—翌年2月末。建议在饲养过程中延长雌、雄赤斑羚合笼时间(10月—翌年3月),并加强合笼期间行为的观察和记录,从而提升繁殖成功率。 The feces of captive red gorals(Naemorhedus baileyi)in Shanghai Zoo were collected and hormones were de⁃tected to estimate the reproductive cycles.Four methods were used to preserve the fecal samples to evaluate the influence of storage methods on hormone detection over time.The results showed that drying and freezing groups showed better ef⁃fects on estradiol detection,and ethanol immersion and freezing group showed better effects on progesterone,and the freezing group was better on testosterone.The antibiotic storage was only effective within 15 days.Taking all methods into account,the freezing preservation effect is the best,ethanol immersion is suitable for short-term sample transportation and storage,and drying method should be an alternative method when samples cannot be frozen.The detection of fecal repro⁃ductive hormone showed that estradiol peaked at early January and whole December 2018,reached a high level from Feb⁃ruary to April 2019,and peaked from October to December 2019.The progesterone had multiple peaks in January,Novem⁃ber and December 2018,January 2019,and remained high level in February and fluctuated from October to the middle of December.Testosterone peaked from November 2018 to February 2019,as well as in October and November 2019.Ac⁃cording to data collected,we predicted that the reproductive period of female N.baileyi starts from October to the end of March of the next year,in which December to January is the peak period of estrous,and the oestrous period of male N.bai⁃leyi is from October to the end of February of the next year.It is suggested that the cage merging could be extended(from October to March of the next year),and the observation and recording of animal behavior during cage merging period should be strengthened,so as to improve the reproductive success.
