摘要
为了减少因盲插而造成子宫的机械损伤和穿孔,提高小鼠子宫灌注的准确率与成功率,连续5 d对雌性小鼠阴道上皮细胞进行H.E染色,确定发情周期,通过具有照明与摄像的仪器辅助引导子宫插管,评估各时期子宫颈口紧张度及子宫插管难度,然后在第5天筛选出子宫插管评价为困难的小鼠,通过可视化引导对其子宫灌注染液,最后处死小鼠,解剖小鼠腹腔,检查子宫染色情况。结果显示,发情期小鼠子宫颈口紧张比例低于其他情期小鼠(P<0.05),发情间期小鼠子宫插管困难比例高于其他情期小鼠(P<0.05);对第5天进行子宫灌注染液的小鼠解剖,显示子宫染色全部成功。可视化引导子宫灌注提供了可靠易行的子宫灌注方法,可减少因盲插或开腹方法所引入的人为因素及误差。
The objective was to reduce the mechanical damage and uterine perforation causing by blind insertion,and improve the accuracy and success rate of uterine perfusion in female mice.H.E staining was performed on the vaginal epithelial cells of female mice for 5 consecutive days to determine the estrous cycle,guided uterine intubation by an instrument with lighting and camera,and evaluate the tension of the cervix and the difficulty of uterine intubation in each period.After 5 days,the mice uterine intubation evaluated as difficult were screened,and the uterus was perfused with dye solution through visual guidance.Finally,the mice were sacrificed and the abdominal cavity was dissected to check the uterine staining.The results showed that the ratio of cervical orifice tension in mice in estrus was lower than that in other estrus(P<0.05),and the proportion of difficult uterine intubation in mice in diestrus was higher than that in other estrus(P<0.05).The anatomical results of the mice injected with the dye solution on the fifth day showed that the uterus staining was all successful.Visual guidance of uterine perfusion provides a reliable and easy uterine perfusion method to reduce the human factors and errors introduced by researchers due to blind insertion or laparotomy.
