Prevalence and Molecular Typing of Rhinoviruses in People with Severe Acute Infection of the Respiratory Tract in Guangdong Province,China,2019–2021

摘要 分析广东省人鼻病毒(Human rhinovirus,HRV)的流行情况和型别分布情况。选取了2019年1月至2021年12月收集的1959份SARI病例的咽拭子标本,采用呼吸道多病原检测和巢式PCR的方法进行HRV的检测和分型,使用SPSS 26.0软件分析HRV的流行分布,使用MEGA11等软件对HRV进行分型和亲缘关系的研究。共收集1959例严重急性呼吸道感染病例(Severe acute respiratory infection,SARI),其中男性为1205例,女性为754例。婴幼儿组阳性检出率最高,随着年龄的增长,鼻病毒的阳性检出率会下降。2020年鼻病毒阳性率仅在6月份有一个高峰期,2021年鼻病毒阳性率在4月份和10月份有两个高峰期。人鼻病毒合并其他病毒检出的病例有25例,检出率为12.31%,与人副流感病毒和人冠状病毒的合并检出最为常见。203份鼻病毒阳性样本共获得114条VP4/VP2序列,分型鉴定结果显示HRV-A型68株,HRV-B型8株,HRV-C型27株,HRV-A和HRV-C是广东省HRV流行的主要型别。婴幼儿是HRV感染的高发人群,HRV-A1和HRV-C15是广东省HRV感染的优势基因型。另外,学校开学导致的学生聚集现象是鼻病毒检出率上升的重要原因之一。 To determine the prevalence and type distribution of human rhinoviruses(HRVs) in Guangdong Province(GP) in China. A total of 1959 throat swabs from severe acute respiratory infection(SARI) cases collected from January 2019 to December 2021 were selected. Respiratory multi-pathogen detection and nested polymerase chain reaction were performed for the detection and typing of HRVs. SPSS 26.0 was used to analyze the prevalence and distribution of HRVs. MEGA11 and other types of software were employed to study HRV typing and phylogenetic relationships. We collected the data for 1959 cases of SARI(1205 males and 754females). HRV-positivity was highest in infants and decreased with age. In 2020, HRV-positivity had only one peak(in June) whereas, in 2021, peaks occurred in April and October. Twenty-five patients had HRV combined with other viruses, percent detection was 12.31%, and the most common cases were combined with human parainfluenza viruses and human coronaviruses. A total of 114 VP4/VP2 sequences were obtained from 203 HRV-positive samples. We detected 68 strains of HRV-A, eight strains of HRV-B, and 27 strains of HRV. Infants and young children carry a high risk of HRV infection. HRV-A1 and HRV-C15 were the dominant genotypes of HRV infection in GP. School meetings led to a marked increase in detection of HRVs.
