摘要 目的 确定赤芍多糖的最佳提取条件。方法 采用苯酚-硫酸法显色,紫外可见分光光度法(UV)在490nm波长处测定当归多糖的吸光度,以赤芍多糖得率为指标,采用单因素和Box-Behnken响应面法优化实验条件。结果 当归多糖的最佳工艺参数为:料液比1∶20.84(g∶m L),回流时间为157min 5s、回流次数3次。结论 最佳提取条件下获得的平均赤芍多糖得率为8.070%,与理论值之间的误差为0.36%,说明将Box-Behnken法应用于水提赤芍多糖工艺的优化是可行的,可用于赤芍多糖的提取。 OBJECTIVE To determine the best extraction conditions of Paeonia lactiflora polysaccharide.METHODS The absorbance radix paeoniae rubra polysaccharide was measured at 490nm by phenol-sulfuric acid chromogenesis and UV spectrophotometry,and the experimental conditions were optimized by single factor and Box-Behnken response surface method using the yield of radix peaoniae rubra polysaccharide as the index.RESULTS The optimal process parameters for the extraction of radix paeoniae rubra polysaccharide were:material-liquid ratio 1∶20.84 g·mL-1,reflux time 157min 5s,and reflux times 3 times.CONCLUSION The average yield of radix paeoniae rubra polysaccharide obtained under the optimal extraction conditions was 8.070%,and the error between it and the theoretical value was 0.36%,indicating that it is feasible to apply the Box-Behnken method to optimize the process of aqueous extraction of radix paeoniae rubra polysaccharide,which can be used for the extraction of radix paeoniae rubra polysaccharide.
