摘要
目的观察缺氧环境中犬内皮集落形成细胞(ECFCs)成血管能力及自噬水平。方法取对数生长期的ECFCs,分为缺氧组和常氧组,缺氧组在缺氧环境中培养,常氧组在常氧环境中培养,比较两组成血管能力(细胞OD值、细胞迁移数、细胞迁移率、管分支数及VEGF mRNA、FGF mRNA、VEGF蛋白、FGF蛋白相对表达量)和自噬水平(细胞LC3 mRNA、P62 mRNA、LC3Ⅱ/Ⅰ、P62蛋白、LC3平均荧光强度、P62平均荧光强度)。结果与常氧组比较,缺氧组培养48、72、96 h的OD值及细胞迁移数、细胞迁移率、管分支数和VEGF mRNA、FGF mRNA、VEGF蛋白、FGF蛋白相对表达量增加(P均<0.05)。与常氧组比较,缺氧组LC3 mRNA、LC3Ⅱ/Ⅰ表达增加及LC3平均荧光强度增强,P62 mRNA、P62蛋白表达减少及P62平均荧光强度减弱(P均<0.05)。结论缺氧环境中犬ECFCs成血管能力增强,自噬水平升高。
Objective To observe the angiogenic ability and autophagy level of canine endothelial colony-forming cells(ECFCs)in hypoxic environment.Methods ECFCs in the logarithmic growth phase was divided into the hypoxic group and normoxic group.Cells in the normoxic group were cultured in normoxic environment,and cells in the hypoxic group were cultured in hypoxic environment.Angiogenic ability(cell OD value,the number of migration cells,cell migra⁃tion rate,tube branch number,and relative mRNA and protein expression levels of VEGF and FGF)and autophagy level(the expression of LC3 mRNA,P62 mRNA,LC3II/I protein and P62 protein,average fluorescence intensity of LC3,and average fluorescence intensity of P62)were compared.Results Compared with the normoxic group,the cell OD values at 48,72,96 h,the number of migration cells,cell migration rate,tube branch number and the relative mRNA and pro⁃tein expression levels of VEGF,FGF in the hypoxia group increased(all P<0.05).Compared with the normoxic group,the expression of LC3 mRNA,LC3II/I protein and the average fluorescence intensity of LC3 increased,while the expres⁃sion of P62 mRNA,P62 protein,and the average fluorescence intensity of P62 decreased in the hypoxia group(all P<0.05).Conclusions The angiogenic ability and autophagy level of ECFCs are enhanced in hypoxic environment.
