多肽水凝胶涂层抗巨噬细胞黏附的研究

Study on the peptide hydrogel coating against macrophage adhesion

摘要 目的研究自组装多肽(RADA)_(4)水凝胶作为载药涂层材料时,抗炎症细胞的黏附作用。方法将(RADA)_(4)水凝胶涂覆于316L SS医用不锈钢表面,对水凝胶微观形态进行圆二色谱和扫描电镜表征,测定其表面亲水性程度。探究(RADA)_(4)水凝胶对RAW264.7细胞黏附、增殖、转化能力的影响;并对其产生的炎症因子进行ELISA检测。结果体外细胞实验表明:(RADA)_(4)水凝胶在短期对小鼠巨噬细胞有抗黏附作用,并显著提高了抗炎因子白细胞介素-10的释放。结论(RADA)_(4)水凝胶作为医用多肽涂层或药物递送系统具有一定程度的抗炎作用。 OBJECTIVE To investigate the role of(RADA)_(4) hydrogel as a drug-carrying coating material in anti-inflammatory cell adhesion.METHODS The(RADA)_(4) hydrogel was coated on the surface of 316L SS medical stainless steel.The microscopic morphology of the hydrogel was characterized by CD,SEM,and the degree of hydrophilicity of the material surface was also determined.The effect of(RADA)_(4) hydrogel coating on the adhesion,proliferation,and transformation ability of RAW264.7 cells was further investigated.The inflammatory factors were determined by ELISA.RESULTS In vitro cellular assays showed that(RADA)_(4) hydrogel had an anti-adhesive effect on mouse macrophages in the short term and significantly increased its release of the anti-inflammatory interleukin-10.CONCLUSION As a medical polypeptide coating or drug-delivery system for medical devices,the(RADA)_(4) hydrogel have some degree of anti-inflammatory effect.
