摘要
目的通过筛选胃癌与正常组织的差异基因,分析并预测治疗胃癌的生物标志物及相关中药。方法利用基因表达数据库(GEO)获取基因芯片,通过GEO2R工具获取差异表达基因(DEGs),进一步通过DAVID数据库对DEGs进行GO和KEGG分析。通过STRING数据库构建蛋白-蛋白互作网络,并筛选出关键基因,分析关键基因mRNA表达和生存曲线。利用Coremine Medical数据库预测作用于关键基因的中药。结果3个基因芯片中共筛选出23个DEGs,其中17个上调基因,6个下调基因。GO分析表明差异基因主要富集在细胞外基质(ECM)、细胞外结构组织、蛋白质细胞外基质等。KEGG分析差异基因主要富集在细胞因子-细胞因子受体相互作用、ECM-受体相互作用、蛋白质消化吸收、cAMP信号通路、PI3K-Akt信号通路等通路。通过CytoHubba分析后得到10个关键基因,其中Ⅰ型胶原α1(COL1A1)和含胶原三螺旋重复序列1(CTHRC1)被视为核心基因,COL1A1和CTHRC1的表达有助于区分肿瘤组织和正常组织,并与生存预后有关。预测出治疗胃癌的潜在中药代表药物有鹿茸、阿胶、党参等(P<0.05)。结论COL1A1和CTHRC1有望成为诊断和治疗胃癌的生物标志物,该研究为治疗胃癌的中药及中成药的开发提供参考方向。
Objective By screening the differential genes between gastric cancer and normal tissues,to analyze and predict the biomarkers and related Chinese medicines for treating gastric cancer.Methods Gene chips were obtained from gene expression database(CGEO),differentially expressed genes(DEGs)were obtained from GEO2R tool,and then the DEGs were analyzed by CO and KECG from DAVID database.Protein-protein interaction network was constructed by STRING database,and key genes were screened out,and the mRNA expression and survival curve of key genes were analyzed.Prediction of traditional Chinese medicine acting on key genes by Coremine Medical database.Results A total of 23 DECs were screened from 3 gene chips,of which 17 genes were up-regulated and 6 genes were down-regulated.CO analysis showed that the differential genes were mainly concentrated in extracellular matrix(ECM),extracellular structure and tissue,protein extracellular matrix,etc.KECG analysis showed that the differential genes were mainly enriched in cytokine-cytokine receptor interaction,ECM-receptor interaction,protein digestion and absorption,cAMP signaling pathway,phosphatidylinositol 3 kinase(PI3K)-protein kinase B(Akt)signaling pathway and so on.Ten key genes were obtained by CytoHubba analysis,among which typeⅠcollagenα1(COL1A1)and collagen-containing triple helix repeat sequence 1(CTHRC1)were regarded as core genes,and the expression of COLIAl and CTHRC1 was helpful to distinguish tumor tissue from normal tissue,which was related to survival and prognosis.It is predicted that the potential representative drugs of traditional Chinese medicine for treating gastric cancer are cartialgenous,asini corii colla and radix codonopsis(P<0.05).Conclusion COL1A1 and CTHRC1 are expected to be biomarkers for the diagnosis and treatment of gastric cancer.This study provides a reference direction for the development of traditional Chinese medicine and Chinese patent medicine for the treatment of gastric cancer.
