摘要
目的:研究桃红四物汤对疱疹后神经痛(PHN)大鼠免疫炎症反应的影响。方法:将60只SD级雄性大鼠按照随机数字表法分为对照组、模型组和观察组,每组20只。其中模型组和观察组均制备PHN大鼠模型。造模成功后观察组予以5 g/(kg·d)桃红四物汤灌胃。比较各项指标的差异。结果:模型组、观察组干预后1 d、7 d、14 d机械痛阈值均低于对照组,且观察组干预后7 d、14 d机械痛阈值均高于模型组(均P<0.05)。模型组及观察组血清及脊髓组织白细胞介素-1β(IL-1β)、IL-4和肿瘤坏死因子-α(TNF-α)水平均高于对照组,且观察组上述水平均低于模型组;模型组及观察组血清及脊髓组织IL-2水平均低于对照组,且观察组血清及脊髓组织IL-2水平高于模型组(均P<0.05)。模型组及观察组CD3^(+)、CD4^(+)及CD4^(+)/CD8^(+)水平均低于对照组,且观察组CD3^(+)、CD4^(+)及CD4^(+)/CD8^(+)水平均高于模型组;模型组和观察组CD8^(+)、TGF-β水平均高于对照组,且观察组CD8^(+)、转化生长因子-β(TGF-β)水平均低于模型组(均P<0.05)。模型组及观察组脊髓组织肝配蛋白B2以及EphB2蛋白表达水平均高于对照组,且观察组脊髓组织肝配蛋白B2以及EphB2蛋白表达水平均低于模型组(均P<0.05)。结论:桃红四物汤应用于PHN大鼠模型中改善免疫炎症反应效果显著,值得重点关注。
Objective:To investigate the effect of Taohong Siwu Decoction on the postherpetic neuralgia(PHN)model in rats.Methods:Sixty male SD rats were divided into a control group,a model group,and a treatment group according to the random number table method,with 20 rats in each group.The PHN model was induced in rats in the model group and the treatment group.After modeling,the rats in the treatment group received Taohong Siwu Decoction at 5 g/(kg·d)by intragastric administration.The differences in various indicators were compared.Results:The mechanical pain thresholds of the model group and the treatment group were lower than those of the control group 1 d,7 d,and 14 d after intervention(P<0.05),and the mechanical pain thresholds of the treatment group were higher than those of the model group 7 d and 14 d after intervention(P<0.05).The levels of IL-1β,IL-4,and TNF-αin serum and spinal cord of the model group and the treatment group were higher than those of the control group(P<0.05),and those of the treatment group were lower than those of the model group(P<0.05).The levels of IL-2 in serum and spinal cord of the model group and the treatment group were lower than that in the control group(P<0.05),and that in the treatment group was higher than that in the model group(P<0.05).The levels of CD3^(+),CD4^(+),and CD4^(+)/CD8^(+)in the model group and the treatment group were lower than those in the control group(P<0.05),and those in the treatment group were higher than those in the model group(P<0.05).CD8^(+)and TGF-βlevels in the model group and the treatment group were higher than those in the control group(P<0.05),and those in the treatment group were lower than those in the model group(P<0.05).The expression levels of EphrinB2 and EphB2 proteins in the spinal cord of the model group and the treatment group were higher than those in the control group(P<0.05),and those in the treatment group were lower than those in the model group(P<0.05).Conclusion:Taohong Siwu Decoction has a remarkable effect on the PHN rat mode
