摘要
研究旨在建立鸡蛋中三甲胺含量的定量分析方法——高效液相色谱-质谱联用法,对比选用5%三氯乙酸水溶液作为提取剂提取鸡蛋中的三甲胺,经冰浴超声萃取、高速离心过滤,再经固相萃取小柱净化,最后通过高效液相色谱-质谱联用法测定三甲胺浓度,并通过测定基质抑制率对检测值进行校准。结果显示:鸡蛋中的三甲胺在0~200 ng·mL^(-1)范围内线性相关,r=0.9994,其基质抑制率为13.36%,方法检出限为7.18μg·kg^(-1)。说明所建立的方法精密度、检出限、稳定性均符合试验要求,可满足鸡蛋中三甲胺含量测定的要求。
The aim of this study was to establish a quantitative method for the determination of trimethylamine contents in eggs by high performance liquid chromatography-mass spectrometry.The 5%trichloroacetic acid aqueous solution was used as the extraction agent to extract trimethylamine from eggs,which was extracted by ultrasonic extraction in ice bath,filtered by high-speed centrifugal filtration,and purified by solid phase extraction column.Finally,the concentration of trimethylamine was determined by high performance liquid chromatography-mass spectrometry,and the detected value was calibrated by measuring the matrix inhibition rate.The results showed the linear correlation of trimethylamine in eggs was 0.9994 in the linear range of 0-200 ng·mL^(-1),the matrix effect was 13.36%,and the detection limit was 7.18μg·kg^(-1).The conclusion showed the precision,detection limit and stability of the method met the test requirements,which could meet the requirements of the determination of trimethylamine contents in eggs.
